Extraction And Separation Of Picroside II And Effect Of Pharmacology

Picroside II is one of most and effective components of the roots of Picrorhiza scrophulariaeflor pennel. Previous studies have showed that picroside II can not only potentiate markedly the nerve growth factor (NGF)-induced neurite outgrowth from PC12 cells, but also enhance the growth of PC12 cells and protect PC12 cells from damage-induced by hydrogen peroxide and hypoxia.In the study, we extracted and separated the picroside II and investigated the mechanism which picroside II protected PC12 cells from damage-induced by Glutamate.In vitro, after being treated with picroside II and Glutamate, the viability of PC12 cells was detected by MTT assays, and the apoptosis rate was detected by Flow Cytometry Analyse and DNA fragmentation analysis. In order to understand the underlying mechanism of the protective effect of picroside II, CDCFH was used to detect the reactive oxygen species (ROS), and the expression level of NF-κB was studied by Western blot. Our results showed picroside II can protect PC12 cells from the death induced by glutamate; and the underlying mechanisms may be involved in the inhibition of ROS formation.In vivo, the Kunming mice were treated with AlCl3 to make AD senile animal model. The mouse water maze test, the analysis of AChE, SOD activities in brains, as well as the pathological observation of the hippocampus were carried out to evaluate the mouse model of AD and the mechanism of picrosides protective effect.As traditional Chinese medicine, Picrorhiza Scrophulariaeflora Pennell would be found new effect in the neurodegenerative diseases.
