Prethrombotic State In Rats After Ischemia-reperfusion Of Limb And It's Intervention Study

Objective The aim of this study was to observe the changes of platelet membrane GPⅡb/Ⅲa and TM on vascular endothelial cell in rats after ischemia-reperfusion of limb and to observe the interventional effects of alprostadil and heparin.To investigate the formation of thromb in rats after ischemia-reperfusion of limb and contrast the intervention effect of alprostadil with that of heparin.Method This study was divided into two parts:in the first part,the rats were divided into two groups:sham operation group(N group);the hind limb ischemia reperfusion group(IR group).An animal model of acute hind limb ischemia-reperfusion was developed by clamping femoral artery and banding hind limb of rats.Changes in the expression of platelet membrane GPⅡb/Ⅲa and TM on vascular endothelial cell in rats after ischemia-reperfusion of limb were observed.In the second part,the rats were divided into four groups:sham operation group(N group);the hind limb ischemia reperfusion group(IR group);the hind limb ischemia reperfusion plus heparin group(IRH group);and the hind limb ischemia reperfusion plus alprostadil group(IRA group).The treatment is ditto,the administration is injection though caudal vein before ischemia.Result(1)The first part:The expression of GPⅡb/Ⅲa of platelet membrane in the IR group was significantly higher than that of the N group(P＜0.05);The expression of TM of vascular endothelial cell in the IR group was significantly lower than that of the N group(P＜0.05);(2) The second part:The expression of GPⅡb/Ⅲa in IRH group and IRA group was significantly lower than that of the IR group(P＜0.05);While there were no significant difference between the IRH group and the IRA group in the expression of GPⅡb/Ⅲa of platelet membrane.The expression of TM in IRH group and IRA group was significantly higher than that of the IR group(P＜0.05);There were no significant difference between the IRH group and the IRA group in the expression of TM.Conclusion Acute hind limb ischemia-reperfusion may induce the formation of prethrombotic state in rats,and early heparin or alprostadil treatment can inhibit this state.But their effective component element is different,the comparision of anticoagulative effect between them need to be studied.
