Objective:Alprostadil is a potent peripheral vasodilator drug that binds to the corresponding receptors on vascular smooth muscle and platelets to exert a role in dilating blood vessels and improving microcirculation;it can improve inositol metabolism and increase Na+-K+-ATPase activity,regulates the activity of adenylate cyclase and phosphodiesterase,promotes the increase of intracellular cyclic adenosine monophosphate,activates a series of protein kinases that depend on intracellular cyclic adenosine monophosphate,and expands blood vessels.Inhibit platelet aggregation,increase blood perfusion,improve nerve nutrition,and thus promote nerve repair[37].The use of alprostadil in the treatment of diabetic peripheral nerve damage has been used for many years abroad,which provides us with a new idea for the treatment of peripheral nerve damage in ischemia-reperfusion.However,little is known about the role of alprostadil in peripheral nerve ischemia-reperfusion injury.The researchers studied the effects of electrophysiological changes in the peripheral nerve injury of alprostadil induced by ischemia-reperfusion injury in rabbits,in order to avoid electrophysiological changes during peripheral ischemia and reperfusion.Methods:Rabbits were used as research subjects to improve the animal model of peripheral nerve ischemia-reperfusion injury.A simple and effective method of modeling was established.Animal models were prepared for use.In the animal model of peripheral nerve ischemia-reperfusion injury,50 healthy New Zealand rabbits were selected and the peripheral nerve ischemic reperfusion injury animal model was selected as the study goal.They were randomly divided into four groups of ABCD,B and C.In group D,arterial clipping was used to block the blood flow at the beginning of the common iliac artery on the rabbit side.At the same time,the internal and external iliac arteries were occluded.In the B2,C2,and D2 groups,they were injected before reperfusion for 1,6,and 12 hours.In alprostadil,B1,C1,and D1 were perfused normally,and after 1 hour of reperfusion,the changes in the electrophysiological and ultrastructural pathology of the phrenic nerve in each group were observed;all indicators were compared with the corresponding control group to explore the alprostadil on the limb The effect of ischemia-reperfusion on the possible mechanism of alprostadil in peripheral nerve ischemia-reperfusion injury was further elaborated,and the possible mechanism of alprostadil in peripheral nerve ischemia-reperfusion injury was further elaborated.SPSS software was used for statistical analysis.Measured data were expressed as mean±standard deviation(χ±S),t test was used;count data were expressed as frequency,and χ2 test was used.P<0.05 was considered statistically significant.result:1)The degree of electrophysiological changes is related to the time of ischemia and reperfusion;2)The electrophysiological changes of the peripheral nerve after ischemia,with the prolongation of ischemic time,the incubation period of ischemia is gradually attenuated,and the action electric force gradually increases;3)Changes in Amp and Lan values at 1 hour after reperfusion of ischemia.Compared to ischemia,Amp was slightly larger and Lan was longer.In ischemia-reperfusion group,Amp decayed in each time group,which was higher than that in ischemia for 1 hour.Lan lengthened and prolonged compared with the control group.After ischemia for 12 hours,alprostadil.Amp and Lan were all slower and lower than ischemia for 6 hours,and they changed significantly after reperfusion.Alprostadil exerted a certain influence on peripheral nerve electrophysiological changes in peripheral nerve ischemia-reperfusion.Conclusion:This experiment confirmed the existence of peripheral nerve ischemia-reperfusion injury in rabbits.The longer the ischemia time,the more obvious the electrophysiological changes.After reperfusion,through 12 hours of dynamic observation,it was seen that within a certain time range,the longer the time after reperfusion,the greater the electrophysiological change.Alprostadil can protect the peripheral nerves after reperfusion.
