Objective: Two cell models were established in vitro to study the antiallergic effects of the natural medicine extracts of Q,and to study the regulative effects of Q on Th1/Th2 balance. Methods: (1) The proliferations of RBL-2H3 cell, splenocyte, and the growth of rat peritoneal mast cells (RPMC) were detected by the method of MTT. (2)The inhibitons of Sodium Cromoglicate and Q onβ-hexosaminidase release from RBL-2H3 cells and RPMC were detected by zymological method. (3) The effects of Dexamethasone and Q on the release of IL-4 and IFN-γfrom mouse splenocyte were detected by ELISA method. Result: (1) In the MTT experiments, the concentration range of Sodium Cromoglicate and Q was 1~100μg/mL , in which the inhibition ratios of RBL-2H3 cells and RPMC proliferation were less than 50%.This concentration range could be used in the zymological experiments. In the mouse splenocyte proliferation experiments, the concentration range of Dexamethasone ,Q3 and Q4 was 1~100μg/mL, in which the inhibition ratios of splenocyte proliferation were less than 50%.This concentration range could be used in the ELISA experiments.(2) Two cell models were established in vitro, the positive drug of Sodium Cromoglicate inhibitedβ-hexosaminidase release from RBL-2H3 cells and RPMC (compare with model group, P<0.01).(3)Utilized the methods of these mast cell models to screen the natural medicine extracts of Q, Q3 and Q4 inhibitedβ-hexosaminidase release from RBL-2H3 cells and RPMC (compare with model group, P<0.01).(4)In the ELISA experiments, Q3 and Q4 down regulated the splenocyte cytokine level of IL-4 and IFN-γ(compare with negative control, P<0.01),which may participate in the regulation of Th1/Th2 balance. Conclusion: the release reactions of different mast cells (RBL-2H3 cell and RPMC) had similarity in the screen of antiallergic medicine, which was discussed in this study. Two cell models were established in vitro to study the antiallergic effects of the natural medicine extracts of Q. The methods of these mast cell models were utilized to screen the natural medicine extracts of Q.Q3 and Q4 inhibitedβ-hexosaminidase release from RBL-2H3 cells and RPMC. Q3 and Q4 down regulated the splenocyte cytokine level of IL-4 and IFN-γ. Q3 and Q4 may participate in the regulation of Th1/Th2 balance.
