Objective To study the effects and pharmacological mechanisms of Tuo Min Zu Fang (TMZF) in treating experimental hypersensitivity typeâ… and typeâ…£in vivo and in vitro. To supply some pharmacological basis for the application of TMZF in treating allergic diseases clinically.Methods (1) In vitro experiment: 1. the effects of Tuo Min Zu Fang to the phagocytosis of macrophages was measured with neutral red method. 2. The proliferative of lymphocytes to ConA was measured with MTT assay.(2) In vivo experiments: 1. We study the effects and pharmacological mechanisms of TMZF in treating experimental hypersensitivity typeâ…£. 1). The proliferative of lymphocytes to ConA was measured with MTT assay. 2). The concentration of IL-2 and IFN-γsecreting by lymphocytes was measured with ELISA. 3). The phagocytic capacity of peritoneal macrophages was measured with phagocytosing chicken erythrocytes method. 2,We study the effects and pharmacological mechanisms of TMZF in treating experimental hypersensitivity typeâ… . The concentration of IL-4 and IFN-γin serum was measured with ELISA.Results (1) TMZF 1. 4g/ml and 2.8g/ml significantly inhibit the phagocytic capacity of peritoneal macrophages as compared with that in the control group (p<0.01). (2) TMZF 1.4g/ml and 2.8g/ml could enhance the lymphocytes proliferation induced with ConA in vitro as compared with that in the control group (p<0.01). (3). TMZF 1.4g/ml and 2.8g/ml could inhibit the lymphocytes proliferation induced with ConA in vivo as compared with that in the control group (p<0.05). (4). TMZF could inhibit the concentration of IL-2 and IFN-γsecreting by lymphocytes as compared with that in the control group (p<0.05). (5) TMZF could inhibit the phagocytic capacity of peritoneal macrophages measuring with phagocytosing chicken erythrocytes method as compared with that in the control group (p<0.01). (6)TMZF could increase the concentration of IFN-γin serum and decrease the concentration of IL-4 in serum as compared with that in the control group (p<0.05).Conclusions (1)It is suggested that TMZF possesses an antagonistic effect on hypersensitivity typeâ… and typeâ…£. (2) TMZF may inhibit the phagocytic capacity of peritoneal macrophages, could inhibit the lymphocytes proliferation induced with ConA in vivo. (3)TMZF may adjust the release of some cytokine, it may be the main mechanisms of TMZF ' effects of anti-hypersensitivity typeâ…£. |
