| Objectives: To establish the auricular phlebitis model of rabbits by intravenous infusion of vinorelbine.To evaluate the protection effect of the new drug chitosan on vinorelbine-induced phlebitis.Methods: Administered vinorelbine intravenously by different doses , diluent volumes,and infusion speed.Observed the difference of the phlebitis happening and analyzed the pathological features of different groups; accordingly established the reasonable phlebitis model induced by vinorelbine.Providing different protection methods (chitosan, MgSO4,and blank)after vinorelbine intravenous infusion,observed the difference of the phlebitis happening and pathological features.Results: It was reasonable to set up the model by infusing the moderate-dose vinorelbine (1.0mg/kg) meanwhile controlling the diluent volume as 10ml and the infusion speed as 0.5ml/min.At the time of 48h after vinorelbine intravenous injection,the chitosan group displayed the more gently pathological damadge than the other groups and meanwhile improved the endothelial cell growth by improving the expression of VEGF. Phlebitis symptoms lasted approximately 40 hours in chitosan group which is more shorter than either MgSO4 or blank group. Conclusions: Chitosan is effective on prevention of the vinorelbine-induced phlebitis owing to it could cut down the poisonous time and strength of vinorelbine. |
