Experimental Research On Compound Chinese Traditional Medicine In Treating Rats Model Of Gastric Precancerous Lesions

Objective To study the effect and the possible mechanisms of compound Chinese traditional medicine in the treatment of gastric precancerous lesions.Method Six-week-old male Wistar rats were used for the experiment.The total of 72 rats were randomly divided into the nomal group (n=18), Other 54 rats were model group. MNNG solution was drunk by the model group freely ,Feeding last for 24 weeks in the model group.At the end of 24 week ,one and three rats were killed in nomal group and model group in order to understand model was successful.At this time ,normal group had 17 rats,other model rats were then randomly divided into model control group(n=15) ,tretinoin group(n=16),CCTM group (n=17).Afterwards rats of CCTM and tretinoin groups were respectively fed in CCTM with decotion and tretinoin for 16 weeks. After 40 weeks, rats of 4 groups were sacrificed .The change of gastric mucosa in 4 groups in histopathology ( including IM,ATP) were observed. The expression of PCNA (proliferation cell nuclear antigen) , P53 protein in gastric mucosa epithelial cell were detected by immunohistochemitry, and cell apoptosis by TUNEL. We calculated the proliferation index and apoptosis index.Results The incidence of intestinal metaplsia (IM) and atypical hyperplasia (ATP) in normal group ,model control group, tretinoin group and CCTM group rats were 11.8% and 5.9%,86.7% and 80.0%,33.3 % and 26.7%,18.8 % and 12.5%, respectively,Compared model control group with nomal group (P<0.05),the model was copied successfully.CCTM can improve IM and ATP of gastric mucosa(P<0.05).Compared tretinoin group with CCTM group,there was a significant difference(P<0.05).CCTM can obviously down-regulate expression of PCNA and P53 protein,which had a significant difference between CCTM group and model control group (P<0.05),but no significant difference between tretinoin group and the model control (P>0.05).They can up-regulate apoptosis index,but there was a significant difference between CCTM group and tretinoin group (P<0.05).Conlusion :CCTM (Wei ruisheng) can improve model rats condtion IM and ATP , reverse gastric precancerous lesions of rats.It can lower expression of mutated P53 and PCNA protein, and accelerate cell apotosis,suppress cell excessive proliferation,recover balance of cell proliferation and apotosis,which may be one of treatment mechanism of gastric precancerous lesions .
