Objective: To observe the influence of Naoling decotion on ethology, hippocampal histomorphology, hemorrheology of vascular dementia rats and expression of nuclear factor kappaB in hippocamal neurons of the vascular dementia rats, and to explore the therapeutical effect on vascular dementia rats and the potential mechanism of action of Naoling decotion .Methods: There were 50 pure line rats and the model of vascular dementia rats was built up by middle cerebral artery occlusion (MCAO), then they were divided into Naoling decotion group, Hydergine group, model group in random,and there were sham operated group and normal group in addition. Each group had 10 rats. After being cured for twenty -eight days,their ability of learning and memory was evaluated by step down test and Morris water maze experiment. The brain tissues were removed to be made frozen sections and stained with HE, and then the hippocampal histomorphology was observed and analyzed by light microscope. The blood was taken for the detection on the indexes correlated to hemorrheology. Expression of NF-kappa B in the hippocampus was measured with immunohistochemical staining.Results: 1. The results of step down test showed that model group presented extension of response time ( P<0.05 ) , decurtation of latency period(P<0.05) and increase of error times(P<0.05) compared with sham-operated group and normal group; compared with model group, Naoling decotion group and Hydergine group presented decurtation of response time(P<0.05), extension of latency period and decrease of error times(P<0.05). Meanwhile, there existed no obvious difference between these two therapy groups.(P>0.05) 2.Compared with normal group, the escaped and latent period of model group was delayed significantly in Morris water maze scoured experiment on delitescence platform (P<0.05). In exploratory experiment, the average pass through times in sham operation group and normal group were more than that of other three ones, and that of the model group was lest, there were significant difference (P<0.05). 3. Under the light microscope, hippocampal CA1 fields of rats in the sham-operated group and normal group exhibited closely and neatly spaced pyramidal cells, and without significant neuron loss;in the hippocampal CA1 fields of rats in the model group sparse and disturbed pyramidal cells ,noticeable neuron loss and neuroglial cell proliferation could be seen. Neuron loss reduced obviously in the hippocampal CA1 pyramidal cell fields of the two therapy group and cell morphology presented relative normal.4. In the blood rheology test, compared with model group,the high shear viscosity,middle shear viscosity,low shear viscosity,plasma viscosity,HCT,Erythrocyte aggregation index and rigidity index were decreased in the Naoling decotion group and Hydergine group,there were significant differences (p <0.05 ) . Compared with Hydergine group,the mean were lower of the high shear viscosity,middle shear viscosity,low shear viscosity,plasma viscosity,HCT,Erythrocyte aggregation index and rigidity index in the Naoling decotion,and there were statistical significance in the middle shear viscosity,low shear viscosity and plasma viscosity (p <0.05) 5.The immunohistochemical integral in the hippocampal CA1 of model rats was 6.16±0.87 and significantly higher than that of shamed-operated and normal rats 0.82±0.23,0.86±0.13 (P<0.05). The integrals in the hippocampal CA1 of rats in Naoling Decotion group and Hydergine group were 4. 02±0.66,3.94±0. 93 and decreased, compared with that of model rats ( P<0.05 ), and there were no significant differences between these two therapy groups (P > 0.05).CONCLUSIONS: 1.Naoling decotion can improve the learning and memory capacity, decrease neuronal loss and better pathomorphological changes in hippocampal CA1 of VD rats, thus it has therapeutic action on vascular dementia rats. 2.Naoling decotion can improve the abnormal indexes of blood rheology and can also improve the hypercoagulabale state of the VD rats' blood.3.Naoling decotion can inhibit the expression of NF-kappa B in hippocamal neurons of VD rats, those partly may be therapeutic mechanism on vascular dementia of Naoling decotion.
