Experimental Studies on the Protective Action of NXF on Vascular Dementia and its Mechanisms of ActionVascular dementia (VD) is an acquired syndrome of intelligence malfunction causedby cerebral vascular diseases including cerebral hemorrhage, ischemia and globalcerebral ischemia and hypoxia. The main clinical symptom is the descending ofmemory and cognitive dysfunction. Some may attend by language barrier, dyskinesia,disorder of spatial resolution and personality changes. In Asia The rate of occurrence isfairly high because of the high rate occurrence of stroke .In China, VD accounts for60~70% dementia and continuously ascends resulted from increasing of theelderly .VD seriously affects patients living quality, at the same time brings a heavyburden to families and the society. In the recent years, people draw a lot of attention onVD, thus both of fundamental research and clinical investigation are developed .There is quite a lot of demonstrations about dementia in Traditional ChineseMedicine. It is considered that VD occurs in the brain and is closely connected with themisadjustment of the heart ,the liver, the spleen and the kidney. Deficiency, stasis,phelm, heat and other factors influence mutually, which are the necessary reasons forthe occurrence of VD. Although there are several types of VD for its differentexpression in clinic, the vacuity of qi and the stasis of blood are the most important andcommon reasons.NXF is mainly composed of the extracts of Radix Ginseng(Renshen), RadixPaeoniae Rubra(Chishao) and Radix Puerariae(Gegen). Nourishing qi and activatingblood is its major efficacy. Our study continues to explore on the protective effects ofNXF on VD models and its mechanisms. The main experimental results are listed asfollows:1 The effects of NXF on the nervous symptom, cerebral infarction area of middlecerebral artery thrombosis (MCAT) ratsAfter MCAT in rats, brain infarction and nervous symptom occurred.NXF(0.24g/kg ,0.12g/kg,0.06g/kg) could reverse the nervous symptom of MCAT rats,decrease the value of nervous symptom assessment at the 6th hour and 24th hour afterMCAT, and decrease infarct size.2 The effects of NXF on the coagulation system of vacuity of qi and stasis of bloodVI 脑欣方对血管性痴呆的防治作用及其机制研究model miceWe put mice into water at a temperature of (10±0.5)℃ and kept them swimming for 5 minutesto build vacuity of qi and stasis of blood model. NXF(0.24g/kg,0.12g/kg) could prolong thecoagulating time(CT)(P<0.01,compared with model), showed dose-effect relation;NXF(0.12g/kg,0.06g/kg) could prolong the activated partial thrombopilastin time(APTT)(P<0.05,P<0.01,compared with model). None of the three doses of NXF show obvious effect onprothrombin time (PT).3 The effects of NXF on blood rheology of acute blood stasis model ratsWe built an acute blood stasis model and found that the whole blood viscosity andplasma viscosity and the erythrocyte aggregation increased, the erythrocytedeformability decrease in the acute blood stasis model rats.NXF(0.24g/kg,0.12g/kg,0.06g/kg) could significantly alleviate the increase of thewhole blood viscosity and plasma viscosity, and could significantly inhibit the increaseof the erythrocyte aggregation, but showed no obvious effect on the decrease oferythrocyte deformability.4 The effects of NXF on learning and memory ability of VD model rats induced bypermanent bilateral occlusion of common carotid artery.Bilateral occlusion of common carotid artery (2 VO) could cause the impairment oflearning and memory ability of VD model rats and neuronal degeneration in brains ofVD model rats stained with H&E method , could decrease the expression of ChAT andBDNF in brains of VD model rats stained with immunohistochemical method.NXF(0.24g/kg,0.12g/kg,0.06g/kg) could improve the impairment of learning andmemory ability, alleviate the neuronal degeneration and...
