| Objectives: To observe the influence of Danshen Root to the expressions of nitricoxide synthase 3 (eNOS) mRNA during acute ischemia reperfusion injury of skeletal musclesin SD rats, and detect the change of nitric oxide synthase (NOS), nitric oxide (NO), ET-1 inblood serum and basic fibroblast growth factor (bFGF) in local skeletal muscle. The studyreveal the mechanism of Danshen Root relieve the disfunction of vessel endothelium duringischemia and reperfusion injury.Methods: Under the anesthesia, the left cremaster muscles of 60 rats weighing 90±10gwere isolated. The vascular clamps were used to completely cut the blood flow of leftcremaster muscles in 66 rats. 6 rats was administered for model identification with bloodvessel opacification. 30 rats were administered intraperitoneally 30 min prior to reperfusionvia Danshen Root served as Danshen Root Group and the rest were administeredintraperitoneally via Normal Sodium (NS) instead served as contrast group. Then, dividingthem into five time points respectively, i.e. 10, 20, 40, 60, 90 min, and 6 rats in each one group.The cremaster muscles at left sides were examined the swelling coefficient at the end of eachtime point. Then the cremaster muscle from the left side was preserved for examination of theexpressions of eNOS mRNA by reverse transcriptase-polymerase chain reaction (RT-PCR)and for examination of the expressions of bFGF. Blood serum was collected from abdominalaorta for examination of NO, NOS and ET-1.Results: The expression of eNOS mRNA in Danshen Root group is higher than that incontrol group, and the difference is very significant (P<0.01) at the time points of 20 and 40min, and at the time point of 60 min the difference is also significant (P<0.05). Comparedwith the control group, the volume of NO, NOS in Danshen Root group is obviously higher atthe time points of 40, 60, 90 min (P<0.01) and 20 min. (P<0.05). However the ET-1 is adverse, which in Danshen Root group is lower than that in control group after time point of10 min (P<0.05). The expression of bFGF is increaseing after the reperfusion, 40 minuteslater the Danshen Root group is obviously more than control one (P<0.01). The test ofswelling coefficien showed the tissue swelling peak is at the point of 40 min, the swellingcoefficien of Danshen Root group is significantly smaller than control group after the timeponit of 20 min (P<0.01).Conclusions:1. There exists severe disfuction of vessel endothelium during ischemia and reperfusioninjury, thus causes ischemia reperfusion injury.2. Danshen Root can increase the expressions of eNOS mRNA in skeletal muscles duringischemia reperfusion injury.3. Danshen Root can stimulus body excretes NOS, NO and inhibit the excretion of ET-1,relieve the disfunction of vessel endothelium during ischemia reperfusion injury.4. Danshen Root can release keletal muscle swelling during the skeletal muscle ischemiareperfusion.5. Danshen Root is a good herb which can stimulate the vessel neogenesis after schemiareperfusion injury.6. The application of Danshen Root will be an effective solution for the skeletal muscleischemia reperfusion in clinic. |
