| Objective To study effective function mechanism of Pilose antler peptide on OA.Method Six New Zealand white rabbits were divided into two groups randomly,each group 4 .The animal model of OA was induced by Hulth's method in animals of experiment group,and the gonarthrotomy was only performed on ones of control group.After 8 weeks,rabbits were sacrificed and isolated the chondrocytes from the OA rabbits bilateral knee joint cartilage then to culture ,so did the control rabbits;Took PAP as investigational drug and took Ossotide Injection as positive control drug. Then determined cell growth basic rule of OA chondrocytes and normal chondrocytes under different drug by Cell counting and drawing growth cure . The proliferation function of chondrocytes was observed by MTT method after treated with PAP. The MMPs'content in ECM of chondrocytes secretion was observed by SDS-PGE.The expression of MMPs mRNA of OA and normal chondrocytes in nucleic acid was examined by RT-PCR method and AGE .Results Osteoarthritis chondrocytes of rabbits cell cycle was shorter than that of normal chondrocytes of rabbits. OA chondrocytes of rabbits cell proliferation was promoted by pilose antler peptide. The variance of MMPs'content in ECM of OA chondrocytes of rabbits secretion was not significant.The level of expression of MMPs mRNA of OA chondrocytes of rabbits in nucleic acid is high.Conclusion Normal differentiation and proliferation of Chondrocyte generates variation in the morbidity process of OA chondrocytes of rabbits .The process of osteoarthritis mesochondrium degradation is possibly attributed to the synergistical consequence of MMPs and other factors. Pilose antler peptide can promote the proliferation of OA chondrocytes of rabbits ,inhibit the overexpression of MMPs of OA chondrocytes.It can possibly play a effective role in the treatment of OA. |
