| Objective To observe the role of H1N1 influenza virus and vaccine in inducing apoptosis of tumor cells and immunoregulation,and to lay the experimentel basis for influenza virus and vaccine as the anti-tumor biological agents. METHOD Detect the effect of H1N1 influenza virus on Hela and B16 cells , influenza vaccine on Hela cells proliferation by MTT method after infecting influeza virus and vaccine; The infected cells were collected to measure apoptosis and cell cycle through flow cytometry. By MTT method ,detecte the effect of H1N1 influenza virus and vaccine on proliferation of mouse splenocytes ;Detecte the content of cytokine in mouse splenocytes supernatant by staining with neutral red and viola crystalline. RESULTS 1.10-1, 10-2, 10-3 the three dilute H1N1 influenza viruses can inhibit the proliferation of tumor cells(P<0.01);2. 10-1, 10-2, 10-3 the three dilute vaccines can obviously inhibit the proliferation of tumor cells, having notable difference compared with control group (t=2.75±0.40,P<0.05);3. 10-1 vaccine can induce apoptosis of tumor cells (apoptosis:58.37%) to contrast (apoptosis:1.45%); 4.difirent dilute H1N1 influenza virus can stimulate the proliferation of mouse splenocytes , having notable difference compared with control group P<0.05(α=0.05);5. 10-1, 10-2, 10-3 the three dilute vaccine can stimulate the proliferation of mouse splenocytes , having statistics significance compared with control groupand (t=2.66±0.289,P<0.05);6. 10-1 dilution influenza virus function mouse splenocytes 24h induces supernatants, after raising the supernatants of ratio dilution to determine activity of IFN-γ, 1:2 degree of dilution group and the VSV virus group havesignificance differences(P <0.01,α= 0.05); 7. 10-1 dilution influenza vaccine stimulates mouse splenocytes and collects supernatants .The result shows that 1:2 time of dilution splenocytes supernatants contains IFNγ, having significant differences (t=3.542, p <0.01);8. In influenza virus and vaccine stimulate splenocytes,of the induction supernate ,IL-2 and the TNF activeness have no significant changes, compared with the control group.CONCLUSION The H1N1 influenza virus and vaccine can inhibit the proliferation of tumor cells and have the anti-tumor effect.The role is achieved through inducing apoptosis of tumor cells , stimulating the proliferation of splenocytes and increasing the externalization of INFγ. |
