Research On Antitumor And Immunoregulation Activities In Mice Of Type â…  Hypersensitivity

Object:Many tumours were infiltrated by eosinophilic granulocyte,which was participated in favourable prognosis of patient with tumour in recent years.In order to explore the role of eosinophils in tumor immunity,we investigated the antitumor effect on mice lung adenocarcinoma cell(Lewis)and immunoregnlation activities of the eosinophils in the eosinophilia-higher mice with S₁₈₀sarcoma.Methods:Lewis were treated with the eosinophils at a diffefent concentrations for 12h-48h.Cell proliferation was evaluated by MTT method.Contains of IL-4 and TNF-a in cultured supernatants for 12h were measured by radioimmunoassay.Mice with sarcoma-₁₈₀were fell into two groups:Tumor group and the Eosinophilia group(5%OVA),which were inoculated subcutaneously in mice of right axil with sarcoma-₁₈₀on the 24day.The contronl group was built at the same time.The tumor inhibition rate was caculated by weighing the weights and the tumor alleosis of mice.At the same time pathomorphological changes of S₁₈₀sarcoma was viewed by hematoxylin and eosin(HE);Proliferations of T lymphocyte and B lymphocyte,cytotocity rate of CTL cell and the function of NK cell and phagocyte were measured by MTT method;Antibody titers in serum was determined by agglutination test;The contains of IL-4 and TNF-a in serum were measured with radioimmunoassay.Results:1.Proliferations of Lewis cells was obviously inhibited by the eosinophils from peritoneal fluid of mice(at cell concentration from 1×10⁴ to 1×10⁶).The anti-tumor activity was gradually enhanced with eosinophil granule variety of dose.2.The eosinophils may inhibit tumor growth in vivo in the eosinophilia-higher mice and the tumor inhibition rate was 20.5%.3.Spleen index was increased obviously in eosinophilia group mice with S₁₈₀sarcoma (P＜0.05).The proliferation of T cell and B cell were strengthen obviously by the eosinophils in vivo in eosinophilia group mice when Con A and LPS existed,respectively (P＜0.01 or P＜0.05),and stimulation index was 2.04±0.14.The activity of CTL cell was heightened by the eosinophils in vivo in mice(P＜0.01),and kill rate of CTL cell was 71.48%.The serum contains of IL-4(1.01±0.22 ng/ml)and TNF-a(1.43±0.08 ng/ml)in eosinophilia group mice were increased.4.Phagocytosis of phagocyte was improved by the eosinophils in eosinophilia group mice (P＜0.01).But NK cell activity was not strengthened obviously by the eosinophils in eosinophilia group mice(P＞0.05).Conclutions:1.The Proliferations of Lewis cell were inhibited obviously by the eosinophilic granulocyte from mice.2.The growth of tumor cell in mice with sarcoma-₁₈₀was repressed and the functions of T lymphocyte,B lymphocyte,CTL cell and phagocytic cell were enhanced by the eosinophils in vivo in eosinophilia group mice,and the serum contains of IL-4 and TNF-a were increased in eosinophilia group mice.So,The growth of tumor may be suppressed by the eosinophilic granulocyte which hightened indirectly immunological function of the mice.
