The treatment based on differentiation of syndromes in Chinese Medicine (CM) has manifested its superiority to some chronic and common diseases. Howerver, the detailed explanation of the etiology and pathology for Ischemic Stroke (IS) based on CM theory and the principle of the differentiated treatment and CM formula are still unclear.1. ObjectiveThis thesis concludes the research proceeding on the treatment of Ischemic Stroke based on"Collateral Theory"and reviews the modern biological and pathological interpretation of"Collateral Theory". According to the hypothesis that the disease, symptom-complex, formula in CM are complex system, this study tends to explore the pharmacological effects of TMP experimentally by a HUVEC model research approach.2. Experimental MethodsWith the model of HUVEC lines, the intervention of Chuanxiongqin (TMP) and normal cells as control group, we adopted the following technique in our in vitro study. The cell proliferation assay, MTT, was examined at 0.5mg/ml, 1.0mg/ml, 2.5mg/ml, 5.0mg/ml, 7.5mg/ml, 10mg/ml, respectively.3. The result of experimental studyIt is found that TMP exhibits significant and different effects(vs normal, P<0.05) on HUVEC proliferation at different concentrations. The results show that significantly promoting cell proliferation(P<0.05) can be observed at low concentration(0.5mg/ml), and obvious inhibition(P<0.001) at high concentration(>5mg/ml), thus the curve of the effect of TMP is non-linear. TMP may exert the protective function of vascular endothelial cell.4. ConclusionSince there are two different properties to be found under the intervention of TMP on HUVEC model, the role of TMP which can be in the range of different doses may exist relative specificity. When treating HUVEC model at low dose, TMP may have potential effectiveness of promoting angiogenesis growth and protecting endothelial cell. We can conclude as it follows:â‘ When treating IS, TMP may play a critical important role in protecting vascular at low dose;â‘¡Chinese drug molecular may exhibit dual regulated role;â‘¢The study identified availability of the treatment of ischemic stroke based on"Collateral Theory"in CM and may contribute to guide the drug screening and development which can protect vascular endothelial cells. Meanwhile, this paper presents the further understanding of the basic concepts, such as"the weakness of the path of collateral","the collaterals damaged by poison evils"about the pathogenesis of IS,"clearing the path of collateral"concerning the treatment of IS.
