ObjectiveTo investigate the mechanisms of Qileng Decoction and the therapeutic method of BVQ(Benifiting Vital Qi), Nourishing Yin(NY) and Activating Blood Circulation (ABC) againstfocal Cerebral ischemia-reperfusion injury from angiogenesis; and vertify that BVENYABC hasa better therapeutic effect than BVEABC for ischemia reperfusion injury compared with that ofthe group of QLDWNY, so as to provide laboratory evidence for clinical use.MethodsThe rats were randomly divided into 4 groups including the sham-operation group, theMCAO (Middle Cerebral Artery Occlusion) group, the group of Qileng Decoction withoutnourishing-Yin herbs(the control group) and the Qileng Decoction group(the treatmentgroup). the MCAO model was established with method of internal carotid artery occlusion bysuture. VEGF and Ang-2 were observed with immunohistochemistry method, and themicrovessel numbcr were detected with CD34 at different repeffusion time of 6h, 12h, 1d, 3d and7d, and the pathology changes was observed at 1d reperfusion time.ResultsThere are few VEGF and Ang-2 positive in the sham-operating group. Both the expression ofVEGF and Ang-2 getting up gradually and reach up to the top at the reperfusion of1d, subsequently, getting down gradually. Except at the reperfusion of 6h, Both the expression ofVEGF and Ang-2 in the treatment group and the control group were higer than that in the MACOgroup at the other time points (P<0.01或0.05), and the difference between the treatment groupand the control group was significant (P<0.01或0.05). the microvesscl number was detectedwith CD34 which was few positive at the reperfusion time of 6h, 12h, 1d, and increaseddramatically at the the reperfusion time of 3d, maintain it to 7d. there was difference bctween thethree groups at the different repeffusion time of 6h, 12h, 1d, 3d, 7d (P<0.01或0.05), andthe difference between the treatment group and the control group was significant (P<0.01或0.05). ConclusionThe protective effects of the Qileng Decoction therapy against cerebralischemia/reperfusion injury might be associated with increasing the positive expressions ofVEGF, Ang-2 and the blood vessel proliferation.Moreover, BVENYABC has a better therapeuticeffect than BVEABC for ischemia reperfusion injury.
