Objective generally the mechanism of longevity is thought the results of multi-facts co-effect corporately; there is not only one contributor. Longevity is more likely the result of mankind adapted to the environment. No doubt among the contributors of longevity, the heredity is the most important and major one. Human gene group plan shows that 99.9% genetic codes of whole DNA are common other than nearly 0.1% of DNA different, exhibiting specificity. So hypothetically, the genetic effects on longevity maybe reside in those differences. Among those differences, Human Leukocyte Antigen (HLA) and Short Tandem Repeat (STR) are the major ones presented as polymorphism. This study choose HLA-A,B and 15 STR allele loci as observation to search for genetic determinants on longevity.Methods Select 18 male nonagenarian in DaLian as observe group. In the control group 50 males are selected from blood donors, age between 18-50, no familial disease.Blood samples are drew venously with EDTA anticoagulant. Genotypes of HLA-A, B loci are used SSO method, first the DNA is extracted, and then PCR are performed. Follows the manipulation manual to perform denature and hybridization, dye with SAPE and wash, finally to interpret the results.STR test with kids (Identifiler PlusTM, ABI Co.), loci to be check are D8S1179,D21S11,D7S820,CSF1PO,D3S1358,THO1,D13S317,D16S539,D2S1338,D19S433,VWA,TPOX,D18S51,D5S818,FGA and Amelogenin.
