Objective: To investigate the genetic association with polymorphism of MBL gene in the patients of Han nationality of southern Jiangsu Province with severe hepatitis B, and the possible associations with clinical manifestations of the disease.Methods: To this purpose, we compared 50 patients with severe hepatitis B ,35patients with chronic hepatitis B and 34 healthy controls. First We extracted genome DNA in whole blood. Then we used Pcr amplificatory and DNA sequencing equipment to detect MBL fragment which included MBL52, 54, 57 alleles. Clinical data were collected and relational analysis was performed.We used ELISA to measure serum concentration of IL-10.AND measured serum concentration of LPS with LAL agglutination.Results: There were no mutations of codons 52 and 57 of the MBL in three groups. Mutation rate of the codon 54 of the MBL is significantly higher in group of severe hepatitis B than of chronic hepatitis B and thecontrol. There was no significantly different between chronic hepatitis B and the control on Mutation rate of the codon 54 of the MBL .The correlation of MBL54 point mutation and A/G has been observed (P<0.05), so as correlation of MBL54 point mutation and concentration of IL-10 (P<0.05).We found that rate of infections of patients of severe hepatitis B in mutant of codon 54 was higher than that in no mutant.Conclusion: 1.Mutation of MBL 54 allele may be one of the important factors that influence onset of the patients with severe hepatitis B. 2. We demonstrated that the patients with severe hepatitis B with mutation of MBL 54 allele were easy to infect bacterium. |
