Zedoary Turmeric Oil (ZTO), a volatile oil, was extracted from the dryrhizome of Curcuma phacocaulis Valeton, Curcuma kwangsiensis orCurcuma phaeocaulis Valeton. Studies on Zedoary Turmeric Oil indicatedthat it had powerful therapeutic effects on many diseases with hypotoxicity.At present, Zedoary Turmeric Oil for injection (Zedoray Turmeric Oil andGlucose Injection) is widely used as a dosage form. Lots of clinicalresearches made sure that Zedoray Turmeric Oil and Glucose Injection hadthe strong therapeutic effects on diseases of respiratory system. At the sametime, the cases on adverse reaction of injection increased in recent years. ?Adverse Drug Reactions Bulletin ? reported that there were 221 clinicalcases on adverse reaction of Zedoray Turmeric Oil and Glucose Injectionfrom January in 1988 to July in 2004. In order to diminish the adversereactions of Zedoary Tumeric Oil for injection, peroral dosage form (softcapsule) was developed. In this study experimenters compared withinjection in terms of antiviral activities in vivo and in vitro to supplyexperimental evidences for dosage form change of Zedoray Turmeric Oil.1 .Comparison of antiviral activities in vitro1) Evaluation of cytotoxicitiesThe cytotoxicities of experimental drugs against MDCK and Hela celllines were examined by the method of quantitative colorimetric MTT. Itwas shown that the TC50 of peroral dosage form and ribavirin agsinst twocell lines were both higher than 1000mg·L-1 . Otherwise, those of injectionfor two cell lines were 50mg·L-1 and 45mg·L-1respectively. Theexperimental data explained that of Zedoary Tumeric Oil peroral dosageform could reduce drug toxicities.2) Comparison of pharmacodynamicsThe results of experiment indicated that the experimental druginhibited pathological changes in two cell lines which infected with RSV,Ad3, CoxB3 and H1N1, and intensity of the contribution was direct ratio todrug dose. In statistics analysis, the conclusion showed that logarithmicdose was positively associated with protect rates for all drugs, ZedoarTumeric Oil peroral dosage form showed a significant dose-effectdependence relation (P<0.01). Compared therapeutic index of experimentaldrugs against four viruses provided, peroral dosage form carried out betterantiviral effects than injection.2 .Comparison of antiviral activities in vivo1) Comparison of peroral dosage form and injection on curing viralpneumonia infected with RSV,Ad3,CoxB3.The results of experiment indicated that Zedoray Turmeric Oilimproved immune functions of mice significantly. Compared effects ofdifferent doses of experimental drugs on inhibiting lung index(LI), highdose group was greatest among these groups and inhibited multiplication ofRSV,Ad3,CoxB3, compared to viral control group, TCID50 reduced 1~2logarithmic doses, effects of high dose group were best among all those ofexperimental drug groups also.2) Comparison of therapeutic efficacies of peroral dosage form andinjectionThe experimental results shown that the mice appeared somesymptoms such as anepithymia, fervescence, asthma, dyspnea, emaciationafter 3~4 days infecting, and some mice with severe symptoms died at the4th day. In the experiment, after 4~8 days infecting, mice of viral controlgroup almost were dead, mortality reached 80%~90%, otherwise the groupwith Zedoray Turmeric Oil, mortality was reduced, survival rate of high,moderate dose group attain 90%, had great significant deviation to viralcontrol group (p<0.005), dose decreased, survival rate reduce concomitantly.In addition, there was a significant deviation among high, moderate dose ofZedoray Turmeric Oil peroral dosage form and viral control group(p<0.001).
