Objective:Screen the differential expressed genes of liver-qi sthenic symptom of complex premenstrual sydrome(PMS ) in macaque brain and construct forward and reverse subtractive cDNA library,in order to form a good platform for systemic study of the change of genetic expression and regulation mechanism,provide a solid foundation for liver-qi sthenic symptom complex of Traditional Chinese Medicine related deep mechanism. Methods:Macaque model of liver-qi sthenic symptom complex of PMS is object, apply the technique of SMART to get the the total full-length cDNA from the brain of macaque model.Apply the technique of SSH in positive and negative directions to acquire the differential expressed genes.Apply the technique of vector recombine to acquire recombinant plasmids.Apply blue/white spot test to screen the positve clones and identify the positve clones through enzyme digesting,then the positve clones are sequenced. Results:Obtain 192 positive clones in forward cDNA subtractive library and 175 positive clones in reverse cDNA subtractive library.The fragments of differential expressed genes are inserted into plamids in 178 and 124 clones.Conclusion:For the first time ,apply the technique of suppression subtractive hybridization to study zangxiang of Traditional Chinese Medicine.Probe into the partial deep mechanism of liver-qi sthenic symptom complex.The forward and reverse subtractive cDNA libraries of macaque brain tissues of liver-qi sthenic symptom complex of PMS are constructed successfully, these libraries may provide a solid foundation for screening and cloning differentially expressed gene of this syndrome.
