The Experiment And Clinical Study Of RuanGanKangXianFang(RGKX) Serum On COL-â… , â…¢ MRNA Expression Of HSC-T6 Cells

[Objective]:To find out the mechanism treated hepatic fibrosis with RGKX by observing the influence of RGKX serum on COL-Ⅰ,Ⅲ mRNA expression of HSC-T6 cells; to observe the clinical curative effect of RGKX and compare the difference between RGKX and FuFangBieJiaRuanGanPian(Co-BJRG) for supplying the evidence for new drugs to cure hepatic fibrosis.[ Method]: experiment and clinical study 1.Experiment studyHSC-T6 cells were exposed in different concentrations of 0.9%N.S,RGKX and Co-BJRG drug blood serum for 48 hours, meanwhile TGF- β 1 was added .Then,the COL-Ⅰ,Ⅲ mRNA levels of HSC-T6 cells were measured by quantitative reverse-transcription polymerase chain reaction(RT-PCR). 2.clinical studySetting up two therapy comparison groups which were RGKX and Co-BJRG, to observe the effect of RGKX and Co-BJRG on the patients' symptoms,blood serum marks and liver functions and compare the clinical effect of the two groups. [Result] : 1.Experiment study5%,10%RGKX and Co-BJRG could restrain the COL-Ⅰ,Ⅲ mRNA expression of HSC-T6 cells(P <0.05或P < 0.01), 20%RGKX couldn't do it. To the COL-Ⅰ, there was no difference between 10%RGKX and Co-BJRG, but Co-BJRG was better than 5%RGKX(P < 0.01); To the COL-Ⅲ, 5% and 10%RGKX was better than Co-BJRG (P <0.05,P <0.01) . 2.clinical studyThe two kinds of drugs can all improve the patients' symptom ,liver function and blood serum marks, the effect was different.
