[Objective] The biosynthesis of aldosterone needs the catalyzation of P-450 carbon 11 β-hydroxynase(P-450c11β) and P-450 cholesterol side-chain cleavage (P-450scc) in adrenal glands.Besides aldosterone,prognenolone is the substrate of both cortisone and sex hormone catalyzed by cytochrome P-450 carbon 17 α -hydroxynase (P-450c17α).in the biosynthesis of steroid hormone in adrenal glands. So the activity of P-450c17αmay affect the contents of aldosterone (ALD) by controlling the contents of the prognenolone in adrenal glands.To probe the mechanism of excess biosynthesis of ALD and the significance of differentiation in molecular level between adrenocortical adenomas(APA) and nodular hyperplasia from patients with primary aldosteronism,using molecular biological technique, we study the expression of cytochrome P-450sccmRNA,P-450c11β mRNA and P-450c17α mRNA in adrenocortical adenomas and nodular hyperplasia from patients with primary aldosteronism.[Methods] Total RNA were extracted from 8 cases of APA tissue sample, 4 nodular adrenal hyperplasia tissue sample,7 normal adrenal gland tissue sample, 1 normal abdominal skeleton muscle tissue sample by single-step method, the expression of P-450c11β mRNA and P-450c17α mRNA was examined by both Northern blot and Dot Blot analysis.The expression of cytochrome P-450scc mRNA was only examined by Northern Blot analysis. |
