Objective:To observe the effects of FML on inhibition of the expression of p22phox mRNA and p47phox mRNA in the NADPH oxidase,and investigate the FML’s function of prevention and treatment on cardiovascular complications of diabetes in diabetes and hyperlipidemia rats.Methods Diabetes and hyperlipidemia rats were used as the object of study.Randomly put 60 SD male rats into 6 groups(n=10 per group),namely,normal group,model group,low dose groups of FML,middle dose group of FML,high dose group of rosiglitazone and simvastatin Combined dose group(western medicine group).The normal group of rats were fed with normal diet,the other group of rats were fed with glucose-fat-rich diet.The low dose of FML group,the middle dose of FML group,the high dose of FML group and the western medicine group were respectively gastric gavaged with 0.4g FML/3ml,0.8g FML/3ml,1.6g FML/3ml,rosiglitazone 0.2mg and simvastatin 0.8mg/3ml,once a day,3ml one time,while the normal and model group respectively gastric gavaged with an equal volume of distilled water,all the rats were put to death after 4 weeks.The deposition of thoracic aortic lipid were evaluated by light microscope after HE staining.Blood glucose in the rats tail vein were detected with blood glucose test paper method.Expression of TG,CH,LDL-C and HDL-C in the blood serum were detected with the special kits;The relative amount expression of p22phox mRNA and p47phox mRNA in thoracic aorta were detected by RT-PCR.Result ①The cardiovascular complications of diabetes rat model was successfully established by 8 weeks’ glucose-fat-rich diet.The variable manifestations of blood glucose,TG,CH,LDL-C increased significantly and the lipid pool formed finally;②Model group rats’ blood glucose,TG,CH and LDL-C were higher than normal group(P<0.01).Blood glucose,TQ CH,LDL-C of middle dose group,high dose group and western medicine group were lower than model group(P<0.05 or P<0.01).Low dose group rats’ blood glucose,TG,CH andLDL-C were lower than those of model group though not to a significant point(P>0.05);③Model group rats’ HDL-C were higher than normal group(P<0.01),The expression of HDL-C in the low,middle and high dose groups of FML had no significant changes(P>0.05),western medicine group increased more significantly than that of the model group(P<0.05);④The relative amount expression of p22phox mRNA in thoracic aorta in model group were higher than normal group(P<0.01).The relative amount expression in high dose group and western medicine group decreased(P<0.05 or P<0.01)when compared with model group,however,there were no significant changes when compared with normal group(P>0.05),and there were no significant differencese between the low dose group or middle dose group and model group(P>0.05).⑤The relative amount express of p47phox mRNA in thoracic aorta in model group were higher than that of normal group(P<0.01),and that in high dose group and western medicine group were descended(P<0.05 or P<0.01)compared with model group.Conclusion ①there are lipid pool formation in thoracic aorta,increased blood glucose,TG,CH,LDL-C,p22phox mRNA and p47phox mRNA expression,overreaction of oxidative stress in the diabetes and hyperlipidemia rats induced by glucose-fat-rich diet.②FML can obviously reduce the diabetes and hyperlipidemia rats’ blood glucose,TG,CH,LDL-C and delay lipid pool formation in thoracic aorta.Its mechanism may be associated with FML’s effect of reducing the relative amount expression of p22phox and p47phox mRNA in thoracic aorta,and inhibition of NADPH oxidase activation,thus to reduce oxidative stress damage.FML contributes to the prevention and treatment of the cardiovascular complications of diabetes. |
