| IntroductionHepatitis C virus ( HCV) is the leading cause of chronic hepatitis C ( HC) , There are one point seven hundred million men who were infected with HCV all over the world, affecting approximately 3% of the worlds population. 10 -20% of patients who had hepatitis C virus infection eliminate virus spontaneous, 80% of patients progress into chronic infection,33% of patients with chronic infection may characterized by a progressing liver injury and result in cirrhosis after 20 -30 years, 15% of them promotes the development of hepatocellular carcinoma (HCC ) , the incidence of a disease and death rate is very high, this diseases do harm to the peoples health severely. The mechanism by which HCV infect people is not known exactly. Many scholars think that immune mechanisms are responsible for HC, not the direct injury, Specificity humeral immunity and cellular immunity play important role in HC. So , its important to research the relationship between serum cytokine levels and HCV viral load and ALT in patients with HCV infection.Material and methods1. Study populationPatients: 43 patients are select from Clinic of No. 1 , No. 2 hospital of China University and Shenyang Infectious Disease hospital. 24 men, 19 women, age arrange 12 - 65, mean 42. 2. All patients were negative for HTV antibody, and other hepatotrophic viral antigens and antibodies. Other possible causes of autoimmune and metabolic diseases were excluded, and hadnt cured with immuno -suppressive drugs and Interferon. Controls: 20 blood donors are selected , 10men,10women.2. Samples collectionWe collect vein blood of patients with HC using BD vacuum tube and separate serum which was used to be measured for ALT and antibody of HCV IgG. Remainder was stored at - 701 used to be measured for viral load and EL -6, IL-12.3. ALT values of HCV IgG + patients were assayed by automatic biochemistry analyzer of HITACHI 7600.4. Quantitate HCV RNA of HCV IgG + patients using RT - PCR of model LightCycler PCR analyzer.5. IL-6,BL-12 ELISA Test Kit were purchased from JingMei company of ShenZhen. PCR Test Kit used only for Roche were purchased from Piji company. Serum samples were assayed for their DL-6, IL-12 content.6. Statistical analysis; We analyse data using SPSS11.0. Analyses of IL-6 , IL-12 in patients with HCV infection compare with healthy adults were done with t - test, association between ALT and HCV RNA was analyzed by Spearman s correlation test.Results1. Relationship between serum IL-6 levels and HCV viral load and ALT in patients with HCV infectionThe levels of serum IL-6 (206. 37pg/ml) in HCV .patients were higher in comparison with healthy adults (11.94pg/ml) ( P <0,01) ; Patients with HCV infection demonstrated a significant correlation between IL-6 and ALT ( r = 0.87,P<0.01) and HCV virus load (r= 0.88, P<0.01).2. Relationship between serum IL-12 levels and HCV viral load and ALT in patients with HCV infection The levels of serum IL-12 (235. 04pg/ml) in HCV patients were higher in comparison with healthy adults (75. 92pg/ml) ( P < 0.01) ; Patients with HCV infection demonstrated a significant correlation between IL-12 and ALT (r = 0.83,P<0.01) and HCV virus load(r= 0.94,P< 0.01).DiscussionThe investigation of HC had lasted for almost 30 years, The mechanism by which HCV infect people is not known exactly. Many scholars think that immune mechanisms are responsible for HC, and cytokines play a major role in the progress of chronic hepatitis C.The infection of HCV may result the levels of serum IL-6, IL-12 in patients HC higher than healthy adults, and the higher IL-6 levels may promote polarization of B lymphocyte and CTL, boost up the expression of perfurin gene; accelerate the synthesization and secretion of acute protein. Higher IL-12 levels may promote polarization and proliferation of B lymphocyte, CTL and NK cell, eliminate HCV, and may result immune trauma.
