Objective To investigate the difference of the transporter associated antigen processing (TAP) gene polymorphism between HAN and SHE nationality of Fujian province, then to elucidate the characteristic of TAP gene frequence distribution of two nationalities.Methods The polymorphism of the alleles at position 333,637 for TAP1 and 379,565, 665 for TAP2 in 150 HAN subjects and 50 SHE subjects of two nationality were detected by the method of PCR-ARMS, the position 651 of TAP2 was detected by the method of PCR-RFLP. The result was analyzed statistically by X2 (chi-square test) to compare the difference between two nationalities. Results1 The genotype The genotype I,D are the main part at position 333, 637 for TAP1 in two nationalities of Fujian province, their frequencies are 74.7%,79.0% and 70%,82% respectively; In HAN nationality, the genotype of V-A-R-T are the major part at position 379,565,651 and 665 for TAP2, their frequencies are 56%^ 85.3?cK 88% and 55% respectively; and in SHE nationality, the genotype of I-A-R-T are the main part at position 379,565,651 and 665 for TAP2, then- frequencies are 58%^ 81%> 91%and 55% respectively. The frequency of genotype 379-1 of TAP2 in SHE nationality is significantly higher than that of TAP2 in HAN nationality (X2 test, P O.05) . There is no significantly difference among the other genotype of TAP 1 and TAP2 in two nationalities.2 The phenotype In the HAN nationality, the phenotype of II, DD are over-presented at position 333, 637 for TAP1, their frequencies are 57.3% and 60.7% respectively; And the phenotype IV, AA, RR, AT are over-presented at position 379, 565, 651 and 665 for TAP2, their frequencies are 58.6%, 72.7%, 76%and 78% respectively. In SHE nationality, the phenotype of IV, DD are over-presented at position 333, 637 for TAP1 , their frequencies are 56% and 66% respectively; And the phenotype IV, AA, RR, AT are over-presented at position 379, 565,651 and 665 for TAP2, their frequencies are 64%, 64%, 82% and 86% respectively. The TAP 1333-IV frequency of SHE nationality is significantly higher than that of HAN nationality (P<0.05). However, the TAP2 379-W frequency of SHE nationality is significantly lower than that of HAN nationality (PO.05) ?3 The haplotype In HAN and SHE nationality of Fujian, the haplotype of TAP1A and ID are over-presented for TAP1. There is no significantly difference of TAP1 between two nationalities. In HAN nationality, the haplotype of TAP2A and 2D are over-presented for TAP2 but the 2G were not detected. In SHE nationality, the haplotype of TAP2A and 2C is over-presented for TAP2. The TAP2B haplotype frequency of HAN nationality is significantly higher than that of SHE nationality(X2 test, PO.001), but the TAP2D of HAN nationality is significantly lower than that of SHE nationality(X2 test,P<0.05).Conclusion In this study, it is shown that there are three significantly differences between two nationality about the TAP gene frequence distribution. First, the genotype frequence of TAP2 379-1 in HAN nationality is significantly higher than that of SHE nationality. Second, the phenotype frequency of TAP2 379-VV in HAN nationality is significantly higher than that of SHE nationality and the TAP1 333-IV is significantly lower than that of SHE nationality. Third, the haplotype frequency of TAP2B in HAN nationality is significantly higher than that of SHE nationality but the TAP2D is significantly lower than that of SHE nationality.
