Research On PAGE Of Salt-Soluble Protein In Genital And Different Seeds Of Storage Time Of Allium Mongolicum Regel.

Allium mongolicum Regel.is the onion Liliaceae Allium xerophyte perennial,as the special environment,A.mongolicum Regel.has been accounted for the proportion of production and consumption minimal,so the study of other crops with a big gap there.To date,research Allium mongolicum Regel.only in the nutrient composition,tissue culture, chromosome and karyotype analysis of the anatomy of flowers and seed germination characteristics of some reports,but the seeds of the Allium mongolicum Regel.of protein research almost zero.Through this experiment,the Allium mongolicum Regel.seeds content of protein, salt-soluble protein extraction conditions,the best salt-soluble protein gel electrophoresis, as well as Allium mongolicum Regel.seeds the entire period of growth and development of salt-soluble protein were studied.I dentified in the Allium mongolicum Regel.seeds protein content,salt-soluble protein extract of the best conditions,the best salt-soluble protein electrophoresis technology and the growth and development during the evolution of Allium mongolicum Regel.seed,to provide technical guidance for its application and further reveal the Allium mongolicum Regel.seeds of salt-soluble protein in seed formation,development,dormancy,germination in the process of the mechanism,which is to improve the importance of biological research and development and utilization of resources that lay a theoretical foundation characteristics.In this experiment for Allium mongolicum Regel.seeds protein and its changes during the development as following conclusions:(1) In the salt-soluble Allium mongolicum Regel.seeds protein content is 8.95g?100g^-1, soluble protein content was 1.77g?l00g^-1.(2) Salt-soluble protein of Allium mongolicum Regel.seeds were extraction with centrifugtion method.The result showed that the optional content of salt were 1%, motionless time were 4 hours,dilution ratio were 1:9.parameters.(3) The optimal conditions for PAGE were as follows.The results showed that 30% H₂O₂5μL,gel concentration 13%,cross-linkages 4%,pH of gel buffer 3.5,and the injecting volumes is 20uL and 25μL.Electrophoresis voltage is high again after the first low-low. (4) Genital reproductive growth of Allium mongolicum Regel.like bud stage,flowering, maturity and dormancy period,the salt-soluble protein expression PAGE is not the same as that with the emergence of each period.(5) Seeds of Allium mongolicum RegeLto enter the storage period,early stage of salt-soluble protein spectra with changes in storage until the latter part of salt-soluble protein bands tended to stabilize,not change.
