Our previous study found that monglicum regel and its' extractive could restrain the activity of fatty acid synthase.upregulate the enzyme activity of hormone sensitive lipase,chang the fatty acid composition and improve the mutton flavor.The aim of this study was to investigate the effects of water soluble extractives from Allium Mongolicum Regel on mRNA expression and promoter region methylation of Lipoprotein Lipase(LPL)and Peroxisome Proliferators Activated Receptory(PPAR?).24 sheep at the age of 5 months were randomly divided into two groups with 12 sheep each.The expriment was lasted for 75 days,including 15 days Preliminary feeding period,and 60 days normal feeding period.Control group were fed basic diets alone,while treatment group were fed basic diets with 0.1%water soluble extractives from Allium Mongolicum Regel.At the end of experiment,6 sheep chosen randomly from each group were slaughtered,and adipose tissue from sheep back was collected in time.The adipose tissue was used for measuring the mRNA expression of LPL and PPAR ?.The results ofelectropherogram of RNA?amplification curve and melting curve agreed with requirements of the experiment.The results showed that gene expression of LPL and PPAR? were significantly(P<0.001)different between control group and treatment group.Compared to control group.the mRNA expression of LPL declined 35%,while PPAR? increased 35%.LPL had four high methylation CpG islands called LPLM1,LPLM2,LPLM3 and LPLM4 respectively.and PPAR? had one high methylation CpG island after sieving by experiments.The methylation ratios of LPL and PPAR? was analyzedafter verification of Sequencing error distribution and the original data base composition distribution.We found that the methylation ratios of 161 dp in LPLM2 and 32dp?57dp in LPLM3 of treatment group were significantly higher than that of control group.32.0%,62.3%,51.9%higher than control group respectively.The average methylation ratios in every position of LPL significantly increased 21.4%.The results of PPAR?had no significantly differences.
