Purpose:From the perspective of emotional pathogenic,explore the pathogenesis of Liver Depression functional dyspepsia.By analyzing the impact of metabolomics expression in the hypothalamus γ- aminobutyric acid and urine samples,to explore the mechanism of electroacupuncture at Zusanli, liver Shu Points of liver depression in functional dyspepsia,and provide scientific theoretical basis for the clinical treatment of liver depression caused by disease.Material and method:Respectively from the observation of behavior science, molecular biology, metabonomics three aspects of inquiry in this experiment.Selected 48 SPF grade healthy Wister rats were randomly divided into four groups, namely control group, model group, electro acupuncture group and sham acupuncture group, 12 rats in each group.In addition to the blank group 12 rats, the other 36 rats modeling, A rat model of depression was made by chronic unpredictable mild stress, the whole 28 days.Modeling for third weeks, the model group was still receiving stimulation; selects Zusanli, liver Shu Points acupuncture stimulation of electroacupuncture group; sham acupuncture group in the stimulation of 1/3, 2/3 from the tip of the tail at acusector, a total of two weeks.Respectively in the one day before the experiment, the experiment for fourteenth days and twenty-eighth days of experimental rats by open field test, sucrose preference test and observe its behavior changes.After the experiment, observation of stomach and duodenum of rats with and whether organic diseases of abdominal anatomy, and the power of gastrointestinal function detection;Extraction of hypothalamic GABA, using immunohistochemistry method to observe its expression;Collection of urine, samples after pretreatment of metabonomics analysis, combined with the use of high performance liquid chromatography- mass spectrometry, observe the change of the situation of small molecular substances in urine samples of peak area.Results:1.Compared with the blank group,the level of score in the open field test in the rats of model group and vertical score decreased significantly(P<0.01).After treatment, the scores of the EA group level and vertical score increased, with statistical significance as compared with the model group(P<0.01); similarly, higher than that in the sham acupuncture group score, and there are differences(P<0.01).2.The results showed that compared with the blank group, the model group rats gastric residual rate increased significantly, intestinal propulsive rate decreased obviously, and had significant difference(P<0.01); after the treatment, the electroacupuncture group and blank control group had no significant difference(P>0.01), compared with model group(P<0.01), and sham acupuncture group comparison of still had significant difference(P<0.01).3.The experimental results show that, the model group compared with the blank group, the rat hypothalamic GABA expression decreased obviously(P<0.01). The expression of EA group gamma aminobutyric acid increased by the treatment, there was statistical significance compared with the model group(P<0.01); the same was higher than that in the sham acupuncture group(P<0.01).4.Research shows, the rats in the model group and blank control group comparison, the urine metabolism in rats of citric acid succinic acid, phenylalanine, alpha- ketoglutaric acid, aspartic acid, myristic acid peak area was significantly different(P<0.05), are decreased; after the treatment, the electroacupuncture group peak area are on the rise there is a difference, and the model group(p<0.05); and sham acupuncture group compared with model group, no obvious changes(P>0.05).Conclusion:1.The establishment of the stagnation of liver Qi type of functional gastrointestinal adverse rat model,through the behavior observation, proved that electroacupuncture Zusanli, liver Shu Points significantly on the treatmentof liver depression functional dyspepsia curative effect.From the molecular level, the detection of hypothalamic GABA, found a positive moderating effect on gamma aminobutyric acid, proof of electroacupuncture at Zusanli, liver Shu Points of gastrointestinal nervous system has good regulating function.2.The establishment of the stagnation of liver Qi type of functional gastrointestinal adverse rat model,found in urine samples of metabolic patterns of the four groups of rats did have different, indicating a successful model, rats in vivo metabolism changes, some metabolites content in different.Combining with the existing data analysis, there may be material for citric acid, succinic acid, phenylalanine, alpha- ketoglutaric acid, aspartic acid, myristic acid, peak area were decreased after EA treatment, the peak area, there is an upward trend.Try looking for endogenous markers in liver depression functional dyspepsia urine metabonomics, to provide the basis for clinical diagnosis and treatment.
