ObjectiveFunctional dyspepsia (FD) is a a clinical syndrome, which with upper abdominal pain, upper abdominal fullness, early satiety, belching, loss of appetite, nausea and vomiting, epigastric discomfort and other symptoms, and via examination can rule out the disease of the gastrointestinal tract; hepatobiliary and pancreas and other organic diseases, the symptoms or repeated seizures, the occurrence of disease were many factors, but the exact cause and mechanism is still unknown.To observe of influence of Weitongxiaopi Decoction on gastric residual volume, plasma motilin (MTL) and serum nitric oxide(NO) in functional dyspepsia Rats models with Liver qi depression-type.Methods1.Experimental groupsThe healthy of 60 Wistar rats were randomly divided into 6 groups:control group, model group, domperidone group, Weitongxiaopi Decoction of high, medium and low-dose group,10 rats in each group.2.Making methodThe Functional Dyspepsia Rats models with Liver qi depression-type were induced by stimulating rats on tails with forceps clip.Everyday (8:00, 11:00,14:00,17:00) rats with rat tail clipped the distal 1/3,30 minutes per time, continuous stimulation for 7 days. Every animal of model group and control group give a certain amount of water and rat food at 8:00am everyday, to survey the residual volume at 8:00 am the next day. 3.Medication MethodsAfter the modeling, fed twice a day by gastrogavage,sustained for 14days, Model group, blank control group,Weitongxiaopi Decoction high, medium, low-dose group, and Domperidone group; normal saline,different concentrations of Weitongxiaopi Decoction (the dose equivalent of adult medicine dosage of medication were 4 times,2 times,1 times, were given 50g/kg/d,25g/kg/d,12.5g/kg/d) and Domperidone were given to six groups. Domperidone group was geived domperidone solution to 3mg/kg/d.4.Experiment targets and detection(1) The general performance of rats:food intake, water intake, activities and so on.(2) Determination of gastric residual volume:After fasting for 12 hours each rats were fed with the semi-solid paste of standard nutritional meal 0.5ml/only,30 minutes later were killed, weigh the wet weight of the stomach first, and then measure the net weight, and the difference was gastric residual volume.(3) Plasma motilin and serum nitric oxide:Abdominal aortic blood collection, subpackage, centrifuge, stored at-70℃refrigerator. Then according to instructions of radio immunoassay to assay motilin (MTL) and NO content.(4) Gastric tissue slice:A portion of stomach tissue, dehydrated, embedded in paraffin, sliced is made, then HE staining and observed under the microscope.5.Statistical MethodsData were mean±standard deviation (x±s) that the use of SPSS11.5 software analysis, the groups were compared with t test and F test, P<0.05 indicated significant difference.ResultsWeitongxiaopi Decoction can only significantly reduced gastric residual volume (P<0.05), middle dose group more effective than high-and low-dose group (P<0.05). Weitongxiaopi Decoction of high, medium-dose group can improve the level of MTL, each dose group were lower NO levels (P<0.05). ConclusionWeitongxiaopi Decoction can reduce gastric residual volume,the secretion of serum nitric oxide and increase the secretion of plasma Gastrin, and improve the gastrointestinal motility of the Functional Dyspepsia Rats models with Liver qi depression-type, the best dose group was medium-dose group.
