For providing the research basis for the prevention of traditional Chinese medicine to treat cerical cancer.Methods:1 Clinical research:The application of the patients of 30 cases of cervical HPV infection protect women Kang Shuan treatment, menstrual clean 2-5 days, every night before going to bed after cleaning the vulva, will protect women Kang Shuan deep in the vagina dome,2 PCS/night. Each menstrual cycle use 20 days, menstrual drug withdrawal, post 3 menstrual cycle is 1 treatment course, stop drug review after one month. Compared with before treatment, analysis of HPV negative rate and clinical symptom score changes before and after the treatment,preserved woman Kang Shuan to evaluate the clinical efficacy of treatment of cervical HPV infection.2 Experimental study2.1 Measurement of cell culture and cell growth curve:24h cell culture after inoculation, Zedoary turmeric oil were added to SiHa cells, HeLa cells and C33a cells of each group cells (0.001-lmg/ml) and the same volume of blank solvent (DMSO). Each cell set up 3 parallel group, and blank control. The effect of MTS method to detect the drug on cell proliferation. The inhibition rate was calculated and selected the best prescription dose;2.2 Detect E6 mRNA expression in cells:In different concentration of zedoary turmeric oil cells after treatment,extracting total RNA.The relative expression was detected by Realtime-PCR HPV E6 mRNA in HPV16 Siha positive cells and HPV18 positive cells in Hela content difference.Results:1 Clinical study:After 30 patients were treated by bao fu Kang Shuan, HPV overcast rate was 43.3%;before and after treatment of clinical symptoms integral by itself after matching t test,t=8.595,P=0,the difference was statistically significant,illustrating Bao fu Kang Shuan that not only can effectively eliminate cervical HPV infection, but also can significantly improve the overall situation in patients with cervical HPV infection.2 With different concentration of zedoary turmeric oil on cervical cancer SiHa cells, HeLa cells and C33a cells, results show the zedoary turmeric oil can inhibit three kinds of cervical cancer cell growth in 0.1mg/ml, in 0.1mg/ml the following can significantly inhibit the growth of cervicalcancer cells. Realtime-PCR is used to display the detection results, before and after different concentrationof zedoary turmeric oil HPV16 E6 mRNA in Siha cells relative expression levels of the P values were 0.1420,0.1216,0.0820,0.0734, 0.0405, HPV18 E6 mRNA relative expression in Hela cells. P values were 0.1612,0.1327, 0.0915,0.0632,0.0312, after statistical analysis, the difference was statistically significant, P < 0.05.Conclusion1 According to the literatures, cervical HPV infection has certain healing rate. The clinicalresearch results show that Baofukang suppository in the treatment of cervical HPV infection in clinical efficacy. Baofukang suppository in the treatment of cervical high-risk HPV infection can not only improve the conversion rate, but also can improve the overall status of the patients, it is worthy of clinical application.2 Zedoary oil is above 0.1mg/ml can inhibit the proliferation of cervical cancer, with a dose-dependent effect, the effect may be achieved by inhibiting transcription of HPV 16 and 18 E6 mRNA. |
