Objective:To study the reverse effect of zedoary turmeric oil serum on multidrug-resistant stomach cancer cell line SGC7901/CDDP and explore the mechanism preliminarily,which may provide experimental base for developing zedoary turmeric oil for reverse drug in treating stomach cancer resistant to multidrug.Methods:1. Serum pharmacology method was used to prepare zedoary turmeric oil seru.2. Inhibiting effect of zedoary turmeric oil on the growth of SGC7901/CDDP cell were evaluated by MTT assay.3. MTT assay was used to detect the changes of inhibiting effect of zedoary turmeric oil on the growth of SGC7901/CDDP cell and calculate its resistance reversal folds.4. Effect of zedoary turmeric oil on the expression of the multidrug-resistant mdr1gene of SGC7901/CDDP cell was detected by RT-PCR method.5. Effect of zedoary turmeric oil on the expression of the multidrug-resistant P-gp protein of SGC7901/CDDP cell was detected by western blot.ResuLts:20%zedoary turmeric oil high-dose serumã€10%zedoary turmeric oil high-dose serumã€20%zedoary turmeric oil medium-dose serumã€10%zedoary turmeric oil medium-dose serum can reverse the multi-drug resistance of the SGC7901/CDDP cell, with the reversal multiple folds respectively as6.06ã€4.27ã€1.45ã€1.3; but it had no inhibiting effect on the expression of the multidrug-resistant mdr1gene and P-gp protein of SGC7901/CDDP cellConcLusion:Zedoary turmeric oil may reverse the multidrug-resistant stomach cancer cell line SGC7901/CDDP externally.The reverse effects of zedoary turmeric oil high-dose serum groups were more obvious than those of medium-dose and low-dose serum groups,which indicated that increasing the dose of animal drug could enhance reverse effect,but it may cause the side effect of gastrointestinal bleeding.The best dose was still needed to explore by experiments.This experiment couldn’t prove inhibiting the expression of the multidrug-resistant mdrl gene and P-gp protein was the typical mechanisms of tumor resistance, which indicated there might be other reverse ways to be explored. |
