Objective:To observe the Hushe-Liyan Prescription of antimicrobial, anti-inflammatory and analgesic actions,and to investigate the effect of this Prescription on the blood, PGE2, IL-1β,IL-6 and TNF-a in experimental damp-heat type pharyngitis rat serum and futher explore mechanism.Methods:1. The inhibitory effect of Hushe-Liyan Prescription:The Hushe-Liyan Prescription in vivo and in vitro Antimicrobial test, as follows:(1) in vitro Inhibitory effect:Test the tested strains of the antibacterial activity, The minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) of Hushe-Liyan Prescription by The plate assay and the standard tube double dilution method. (2) Inhibitory effect in vivo:experiments with mice injected with pre-determined concentrations of Staphylococcus aureus, observed Hushe-Liyan Prescription protect of Staphylococcus aureus infection in mice.2. the anti-inflammatory and analgesic actions of Hushe-Liyan Prescription:Models of writhing test in mice and hot plate in mice were adopted to observe the Hushe-Liyan Prescription of analgesic actions, models of xylene lead to ear swelling of mice and carrageenin lead to Rat toes swelling to observe the Hushe-Liyan Prescription of anti-inflammatory actions.3. within the eating disorders caused by wet environment, improving air temperature and humidity, plus a simulated exogenous Salmonella typhimurium infection in hot and humid Evils, and use 15% ammonia water hot and humid local stimulation of rat pharyngitis (Disease model) model, biochemical detection routine blood test, IL-1β,IL-6 and TNF-ain rat serum, PGE2 changes.Results:In vitro Antimicrobial experiments, Hushe-Liyan Prescription Inhibition of Staphylococcus aureus, streptococcus and pneumococcus, beta maximum inhibition zone diameters of 15mm,22mm,17 mm; minimum inhibitory concentration (MIC) was 200mg/ml,25mg/ml,100 mg/ml; minimum bactericidal concentration (MBC) are 500mg/ml, 50mg/ml,200mg/ml, on the Antimicrobial effect of E. coli and Candida albicans was not obvious; in vivo Antimicrobial experiments, From the average survival time of mice in each group to see, compared with the control group the differences were significant (P<0.01), that Hushe-Liyan Prescription Treatment of radiation administered in vivo to Staphylococcus aureus infection in mice are protected effect. Anti-inflammatory analgesic in the experiment, the Hushe-Liyan Prescription can significantly reduce the xylene induced ear edema in mice and carrageenan induced rat paw edema, and reduce the number of writhing induced by acetic acid, enhance pain threshold of mice. Antipyretic and anti-inflammatory mechanism in the experiment, the Hushe-Liyan Prescription has antipyretic effect of each dose group, rats of each dose in the blood leukocytes, granulocytes and lymphocytes were significantly lower than the model group, and can PGE2 significantly inhibited the release of high-dose group in which the most significant; with the model control group, the Hushe-Liyan Prescription high, medium and low dose group serum IL-1(3, IL-6 and TNF-a levels Significantly lower, the difference was statistically significant (P<0.01).Conclusion:the Hushe-Liyan Prescription have Relatively good antibacterial, anti-inflammatory, analgesic and antipyretic effects, and through reduced Routine blood test leukocytes, granulocytes and lymphocytes, Pharyngeal tissue PGE2 and serum IL-1β, IL-6 and TNF-αlevel to treat damp-heat in the role of pharyngitis.
