The drug property theory of Chinese Medicine is the essential components of Chinese Medicine System,the core of Chinese Medicine Theory system and the foundation for guiding clinical medication of Chinese Medicine.The nature and flavor including four kinds of nature and five kinds of flavor is the basis of drug property theory and the most valuable parts.On the basis of the hypothesis proposed of my tutor in this dissertation:"The hypothesis of scientific connotation on Chinese herbal medicine flavor and nature".The research team carried out theoretical research on the medicinal properties of Chinese herbal medicine for the efficacy of alleviating water.Anemarrhena asphodeloides is the drug that it’ s efficacy of alleviating water retention has been researched here and the research object in this dissertation.Anemarrhena asphodeloides’ nature is cold and its flavor is bitter and sweet,and it belonged to lung,stomach and kidney viscera in Chinese Medicine,furthermore,the function of Anemarrhena asphodeloides is clearing heat,purging fire,nourishing Yin and moistening dryness.Bitter and sweet medicine efficacy of Anemarrhena asphodeloides is taken separately in classical prescription of Baihu Decoction and Zhibaidihuang Pill,moreover,the flavor components and flavor of Rhizoma Anemarrhenae split components being combined to study is carried out to verify what the nature and flavor of split components belong to,clarify its effect and mechanism,which lays the theoretical foundation for the clinical fields.The research on the model of separation and combination of Chinese medicinal herbs’ nature and flavor should be improved further,in order to enrich the theories of Chinese medicinal herbs’ nature and flavor,and provide important reference for clinical application of drugs and development of new drugs.1.Literature ResearchDevelopment of Chinese drug property theory,research status of the main chemical composition and pharmacological action of Rhizoma Anemarrhenae,the classic Baihu Decoction and Zhibaidihuang Pill of prescription compatibility is discussed and main pharmacological and clinical effects were reviewed,the flavor of composability research development has laid a solid foundation for literature.Especially in doctor compatibility,summed up in the Baihu Decoction,the bitter flavor for clearing heat and purging fire effect and anemarrhena asphodeloides in Zhibaidihuang Pill with the effect of nourishing yin and tonifying kidney,its flavor’combination study provides a theoretical basis.2.Chemical characterization study on the material basis of Rhizoma Anemarrhenae’s nature and flavorEstablished in this paper,based on the early stage of the group split method based material base of Anemarrhena asphodeloides was split into four components,namely the polysaccharide components,components of ethyl acetate,saponin constituents and the rest of the components;The methods of HPLC-ELSD and UPLC-MS were used to analyze the chemical characteristics and verify their non-intersections.By silica gel,ODS,Sephadex column chromatography method and HPLC technology such as LH-20,the separation of ethyl acetate part a and part saponins of purification,a total of 24 compounds were isolated,according to the physical and chemical analysis,1D and 2D-NMR spectroscopy and mass spectrometry testing means such as a comprehensive analytical(guide)set of the chemical composition of the structure.The structure of 22 compounds was identified,including 13 saponins,6 flavones and 3 other compounds.3.Combination study of split group on Rhizoma Anemarrhenae’s nature and flavor in Baihu Decoction(1)Study on antipyretic effect of split components of Anemarrhena flavor in Baihu DecoctionIn order to reflect the theory of traditional Chinese medicine in compound Baihu Decoction of Rhizoma Anemarrhenae bitter efficacy as the research object,select the thermal model and the main clinical application,namely four thermal model,2,4 and two nitrophenol,dried yeast,lipopolysaccharide,bacterial endotoxint,study of flavor components,flavor of Rhizoma Anemarrhenae split components instead of the combination of Anemarrhena in Baihu Docotion,the lack of Anemarrhena baihutang group,bitter component(saponin component+polysaccharides+other components)+Anemarrhenae baihutang group,sugariness missing components(ethyl acetate layer)+baihutang,lack of Anemarrhenae polysaccharide fractions+lack of Baihu Decoction,Anemarrhena saponin fraction,the lack of baihutang+Anemarrhenae component missing white tiger Soup+Anemarrhenae antipyretic effect,the results showed that:in the experiment of pharmacology reflected in Baihu Decoction of Rhizoma Anemarrhenae bitter,bitter flavor is an effective component group of Baihu Decoction of Rhizoma Anemarrhenae antipyretic,can replace the raw materials and corresponding compatibility of Rhizoma Anemarrhenae.Split group with bitter flavor of antipyretic effect of saponin and polysaccharide,which play a better role of cooling,the remaining group also has curative effect,but the treatment effect is lower than that of bitter components,further validation of Anemarrhena taste split components belonging,that can be combined the taste of the split group,confirmed the theory of the scientific connotation hypothesis of Chinese herbal flavor.(2)Mechanism study of split group on Rhizoma Anemarrhenae’s nature and flavor in Baihu DecoctionIn Baihu Decoction of Rhizoma Anemarrhenae flavor split component research on combination line four thermal experiment,observe the heating factor is TNF-α,IL-6,cAMP,IL-1β,PGE2 and cAMP/cGMP in the blood and hypothalamus inhibition,and the expression in parts of the lung meridian TNF-α,IL-6,IL-1β factor,found and taste of Rhizoma Anemarrhenae split components of multi-channel and multi targets play antipyretic effect,models of different drug groups the inhibitory factor of different the inherent advantages of the compound of Rhizoma Anemarrhenae,provide an important basis for the scientific flavor of the combination of Chinese medicine and property theory hypothesis(3)Combination study of split group on Rhizoma Anemarrhenae’s nature and flavor in Baihu DecoctionPrototype components and metabolites in the blood detected by UPLC-MS sugariness compound group and bitter compound group after administration,found 1 prototype components,The of methods PCA and OPLS-DA were analyzed before and after administration of compound bitter and bitter compound of blood and urine metabolites in the blood profile,metabolism,analysis method of PCA and OPLS-DA effect of Rhizoma Anemarrhenae sweet components and bitter components of metabolic differences,two distinct effects on Anemarrhena’s nature and flavor in the treatment of nephritis in Zhibaidihuang PillStudy on the flavor components of Rhizoma Anemarrhenae combined therapeutic effect of Zhibaidihuang Pill on nephritis,6 biochemical and histological observation of pathological changes of flavor components of Rhizoma Anemarrhenae split treatment,the results showed that:in the experiment of pharmacology nephritis antipyretic and sugariness Compound Rhizoma Anemarrhenae bitter compound group Anemarrhena components in Baihu Docotion in the Zhibaidihuang Pill in the treatment based on characteristics of TCM syndrome differentiation,to provide a scientific basis for exploring the combination of prescription for clinical use and provide new ideas for further improvement of traditional Chinese medicine the theory of compatibility.
