| Prat 1 The inhibition effect of Yanghe decoction on Yang deficiency breast cancerObjectives: Observe the inhibition effect of Yanghe decoction on 4T1 breast cancer cells and subcutaneous tumor of mice.Methods: Vitro studies: 4T1 breast cancer cells were treated with medicated serum of Yanghe decoction,observe the inhibition effect of Yanghe decoction on 4T1 breast cancer cells.Vivo studies: The mouse model of Yang deficiency breast cancer was established by adenine irrigation,establish the animal model of Yang deficiency breast cancer by hypodermic inoculation.The intervention was carried out with Yanghe decoction and cyclophosphamide.Observe the spleen weight,tumor weight and the inhibition rate.Reuslts:1.The inhibition effect of Yanghe decoction on 4T1 breast cancer cells showed time-dependent.2.28 days of adenine irrigation,the mice showed Yang deficiency signs of cowering,chills,lethargy,and defecation,the expression of T3、T4 and ACTH down-regulated.3.The spleen of the three treatment groups was less than that of the model group(P<0.05),the spleen weight of the combination group was smaller than that of the other two groups(P<0.05).Tumor weight of the Yanghe decoction+ Yang deficiency group and the combination group less than the model group(P<0.05 or P<0.01).Conclusion: The Yanghe decoction shows great inhibition effect on 4T1 breast cancer cells and subcutaneous tumor of micePart 2 The mechanism of Yanghe decoction against Yang deficiency breast cancerObjectives: Explore the mechanism of Yanghe decoction against Yang deficiency breast cancerMethods:1.4T1 breast cancer cells were treated with medicated serum of Yanghe decoction,ELISA detect the expression of VEGF and PGE2.2.flow cytometry was used to detect the content of MDSC,NKT cells and T cell subgroups in spleen of mice.ELISA detect the expression of IL-6、TGF-β and IFN-γ,RT-PCR/DHE detect the expression of i NOS、ARG-1 and ROS,Western bolt detect the expression of stat1、p-stat1、stat3、p-stat3.Results:1.the expression of VEGF and PGE2 down-regulated.2.MDSC and NKT cell contents of treatment group were lower than the model group(P<0.01).The increase of CD3+CD4+T cells of treatment group was obvious(P<0.01),the changes of CD3+CD8+T cells were not obvious(P>0.05).3.The treatment group was significantly up-regulated in expression of IFN-γ(P<0.01),Yanghe decoction+ Yang deficiency group higher than Yanghe decoction + breast cancer group(P<0.05),The treatment group were lower than model group in IL-6 and TGF-β(P<0.05 or P<0.01).The treatment group were lower than model group in i NOS、ARG-1 and ROS(p<0.01).4.The up-regulate of p-stat1/stat1 of treatment group was obvious(P<0.05 or P<0.01),The down-regulate of p-stat3/stat3 of treatment group was obvious(P<0.01),no statistical significance between treatment groups(P>0.05).Conclusion:1.Medicated serum of Yanghe decoction can effectively inhibit the proliferation of 4T1 breast cancer cells.2.Yanghe decoction play an anti-tumor role by inhibiting the generation and activation of MDSC,the main mechanism is down-regulate the expression of VEGF、PGE2、IL-6、TGF-β to inactive the path of stat3.Meanwhile,the expression of i NOS,ARG-1 and ROS of downstream products of MDSC were down-regulated too,make MDSC lose immunosuppressive function.3.Yanghe decoction activate STAT1 by increasing the expression of IFN-γ to enhance the body’s immune response,remodeling immune function by increase the expression of CD3+CD4+T cells. |
