Objective:It' s very common that the cartilage defects in clinic, because of trauma and all kinds of diseases. Due to the limit of cartilage spontaneous regenerative capability, there is no effective treatment in clinic at present. It gets hopeful that the repairing of cartilage defects, because of the development of tissue-engineering and molecular biology. The study of tissue-engineered cartilage is the hotspot of tissue-engineering. The artificial cartilages have been cultured, such as finger articular cartilage, temporal-mandible articular cartilage,auricular cartilage and nasal cartilage, which were similar to normal cartilage structure. It is very difficult to realize reasonable interaction among seeding cell, growing factors and cellular scaffolds, so there is no satisfied method that can product tissue-engineering cartilage to repair cartilage defects at present. In this experiment, we explored the feasibility of producting neocartilage tissue, using allogenic chondrocytes as seeding cells, artificial absorbable scaffold—polylactic acid(PLA) as matrices, perhaps it is helpful for clinical repairing cartilage defects. Method:The cartilage tissue was obtained from the neoborn new Zealand rabbits(born 3-5 days), under sterile conditions, the four limbs of rabbits were dissected, cartilage fragments were sharply curetted off the articular surface of four limbs. Washing with PBS solution, then cartilage fragments were cut into 1×1mm pieces, digested by II collagenase, centrifuged. At last we obtained the isolated chondrocytes. The isolated chondrocytes proliferated in vitro, the culture medium(RPMI1640) supplemented with fetal calf serum(FCS), antibiotics, vitamin C. After four generations, the chondrocytes were collected.
