| Ulcerative colitis (UC), caused by indefinite factor, is a chronic nonspecificinflammatory intestine disease. It manifests itself as diarrhea with bloody purulentstools, abdominal pain and tenesmus. Its pathological characteristics are inflammationand ulcer of colon. To study the functional mechanism of variant sinisan to treat UC,we have UC and sinisan studied in theory and experiment in the following two parts. The study in theory 1.The status of UC in modern medicine The summary of the mechanism of UC, clinical treatment and model of animal inmodern medicine are summed up thoroughly, especially the mechanism of UC inimmunology. The outline of study of UC in modern medicine is mastered. 2.The status of UC in Traditional Chinese medicine(TCM) It is rounded up thoroughly about the knowledge of name of disease of UC, causeof disease and pathogenesis, the status of treatment and experimental research in TCM.We put up with our viewpoint that is impediment in the functional activities of Qi isthe fundamental pathogenesis of UC and has during UC by the main clinicalmanifestation. 3. The status of the study of sinisan It is stated about the theoretical contending of sinisan, the meaning of content ofsinisan, laws of clinical use and the summary of experimental study of sinisan. Weconcluded that the effect of sinisan regulating the fundamental activities of Qiconforms to the fundamental pathogenesis of UC. The study in experiment 1. Methods The healthy male Wister rats is divided 6 groups randomly, which is individuallynormal group, model group, 3 variant sinisan treatment group included large dosegroup, medium and little dose group, SASP treatment group. The UC rats model isperformed by TNBS/Alcohol Solution(100mg/kgTNBS + 50%Alcohol Solution0.25ml) enema and ties. The all groups are given by pouring into stomach individuallyChinese medicine, western medicine and 0.9% NaCl solution according to theequivalent of person to rats. It performs 1 times/day for 3 weeks. After 3 weeks, wecollect blood and the tissue of colon of the rats. Then the following items isobserved:(1)Observing the rats' general condition by eye before killed it.(2)Observingpathological formation of mucous membrane of colon dyed by HE by opticalmicroscope.(3)Measuring the content of MPO, SOD, MDA in mucous membrane ofcolon. (4) Measuring the content of IL-4, IL-10, TXB2, 6-Keto-PGF1a in serum. (5)Measuring the expression of ICAM-1mRNA by RT-PCR method. Results General conditions: The rats' general conditions of normal group and all treatmentgroups after 3 weeks' treatment are better than that of model group. Observation by optical microscope: The structure of colon in normal group isclear and no inflammation. The structure of colon in model group show obviousinflammation and even forms severe ulcer. There is inflammation' improvement in alltreatment group. Content of MPO: MPO content of model group show a distinct increase and issignificant (P<0.01) compared to normal group. The content of MPO every groups ofvariant sinisan is far lower than that of model group (P<0.01). Content of MDA and SOD: The Content of MDA of all groups of variant sinisanhas a obvious decrease than that of model group (P<0.01). The Content of SOD of allgroups of variant sinisan has a obvious increase than that of model group (P<0.01). Content of TXB2: The Content of TXB2 of all groups of variant sinisan has aobvious decrease than that of model group (P<0.01).
