Objective:Research on the immune parameters in mice about Xi-jiao Di-huang Tang and Xiao-fengsan meddle the animal model of chronic dermatitis and eczema, and to analyze its possible mechanism.Methods:(1) Using the experiment of delayed-type hypersensitivity induced mouce model of chronic dermatitis and eczema to determine ear swelling and analysing the ear swelling about Xi-jiao Di-huang Tang and Xiao-fengsan effect on mice.(2) Using FCM (Flow Cytometry) to test the CD4, CD8 levels of spleen cells of mice of the animal models of chronic dermatitis and eczema, and analysing the effects of T cell subsets CD4+, CD8+ and their ratios about Xi-jiao Di-huang Tang and Xiao-fengsan effect on mice.(3) Using the method of double-antibody sandwich ELISA to test mice serum IFN-γ, IL-4 levels of animal models of chronic dermatitis and eczema, and analyzing the effects of immune indicators IFN-y, IL-4 and its ratio about Xi-jiao Di-huang Tang and Xiao-fengsan effect on mice.(4) Using double-antibody sandwich ELISA to determine the mice serum LTB4, LTC4 of chronic dermatitis and eczema animal model, and analysing the effects of of slow reactive substance cysteine leukotriene about Xi-jiao Di-huang Tang and Xiao-fengsan effect on mice.(5) Useing Real-time PCR to detect ear skin CysLTR1, CysLTR2 expression of mouce models of chronic dermatitis and eczema, and analysing Xi-jiao Di-huang Tang and Xiao-fengsan effect on the leukotriene receptor in mice expression.(6) Useing Western-blot to detect ear skin CysLTRl expression of mouce models of chronic dermatitis and eczema, and analysing Xi-jiao Di-huang Tang and Xiao-fengsan effect on the mice's CysLTRl expression.Results:(1) Xi-jiao Di-huang Tang and Xiao-fengsan high-and low-dose has a very significant inhibition on mice ear swelling in delayed-type skin hypersensitivity (P all<0.001).(2) Both the medicine intervention 1,2-group of chronic dermatitis and eczema are able to decrease elevated serum CD4, CD8 expression levels and their ratio of the mouce model of chronic dermatitis and eczema, and there is no significant difference between groups 1,2.(3) Both the medicine intervention 1,2-group of chronic dermatitis and eczema are able to regulate mouse serum IFN-r, IL-4 as well as its IFN-r/IL-4 ratio, manifested as decrease the elevated serum IFN-r levels and increase the reduced serum IL-4 levels, furtheremore, it has an decrease effect on the elevated IFN-r/IL-4 ratio, so that the ratio shift to equilibrium.(4) Xi-jiao Di-huang Tang and Xiao-fengsan has a certain extent on Serum LTB4, LTC4 levels in mouse model of chronic dermatitis and eczema.(5) Both the medicine intervention 1,2-group of chronic dermatitis and eczema have significantly role of lower relative CysLTRl mRNA expression levels, and there is no significant difference between 1,2 groups. Both the medicine intervention 1,2-group of chronic dermatitis and eczema have significantly role of lower relative CysLTR2 mRNA expression levels. comparing with medicine intervention 1-group,2-group has an significantly differences in lower relative CysLTR2 mRNA expression levels.(6) Both the medicine intervention 1,2-group of chronic dermatitis and eczema have significantly role of lower relative CysLTR1 expression levels in the ear skin of mice(p<0.001).Conclusions:Xi-jiao Di-huang Tang and Xiao-fengsan high-and low-dose has a very significant inhibition on mice ear swelling in delayed-type skin hypersensitivity. It had showed a significant regulatory role including the CD4 + T cells, CD8 + T cells, as well as their ratio, the TH1, TH2 (IFN-γ, IL-4) as well as their ratio, the LTB4, LTC4 and its receptor under the condition of mice's imbalance immune.
