Objective To.study the effects of Pilose antler polypeptide(PAP) on metabolism of chondrocytes cultured in vitro. Methods This study use advanced modern biology technique, such as cell culturing in vitro, radio immunity, isotope tracing, chemical dying and modern picture treating etc, to study the effects of PAP on chondrocytes division, proliferation, DNA copying â…¡ collagen synzyming, proteoglycan synzyming, chondrocyte apoptosis and the relation between the quantity and effect. To observe the morphology difference between the test group and control group using microscope; To count the chondrocytes in the test group and control group, and understand the effects of PAP on chondrocyte proliferation with MTT; To study the effects of PAP on chondrocyte DNA copying with 3H-TdR tracing; To study the effects of PAP on chondrocyte â…¡ collagen with 3H-proline; To study the effects of PAP on proteoglycan synzyming with chemical dying; To study the effects of PAP on chondrocyte apoptosis with chemical dying and counting. Results In the test group, chondrocyte grows well, which density increase obviously, and which shape like shuttle and triangle; In the test groupwhich concentration i s 0. 08%, chondrocyte proliferate obviously, the quantity of 3H-TdR and 3H-proline is maximum; In the test group, chondrocyte proteolycan synzyming increase obviously; The chondrocyte apoptosis in the test group is less than that in the control group. Conclusion PAP can promote chondrocyte DNA copying, â…¡ collagen and proteoglycan synzyming, and prevent chondrocyte apoptosis, which provide a studying direction to treat OA in clinic.
