Objective:To investigate DAPI labeled adipose-derived stem cells for repair of rabbit degenerated intervertebral disc and the change of type II collagen and proteoglycan.Methods:It’s a animal experiment.Twenty-five female Newland white rabbits on March age, which were randomly divided into two groups, the experimental group is twenty and the control groups is five,were made of degenerated intervertebral disc model,at the same time, cultivate the adipose-derived stem cells.After two weeks of the modeling, the disc of degenerated intervertebral of the experimental group were injected with adipose-derived stem cells,Which were labeled by DAPI,while the he control groups were injected with the equal volume of saline.The specimens of degenerated intervertebral disc of rabbit were obtained after lweeks,2weeks,4week,8weeks of injection.In ever time point,the rabbit,which wer put to death,in eluding the four rabbits in experimental groupand and the one rabbit in control groups.The rabbit were scaned by X and MRI to observing the changes of intervertebral disc before transplantation the adipose-derived stem cells and after8weeks. The specimens were stained with HE and DAPI, scanned by electron microscopy after fixed,and the enzyme linked immunosorbent assay (ELSIA)werw made to detection the concentrationof â… type â… collagen and proteoglycan in order to assessment the ADSCs (adipose-derived stem cells)repair the degenerated intervertebral disc of rabbit and the change of adipose-derived stem cells, type II collagen and proteoglycan.Results:The six of the model of the rabbits is die in twenty five, there is not rabbits to dead for another modeling after complete. ADSCs(adipose-derived stem cells) and physiological saline injection. The eighth weeks of X-ray and MRI showed:in the experimental group, the intervertebral disc height was higher than the control group, the difference was statistically significant. ELSIA detection showed:the concentra-tion of the type II collagen and proteoglycan in the experimental group is higher than in the control group, P<0.05, the difference was statistically significant; Electron microscope, HE staining and DAPI staining show:nucleus pulposus cells was significantly higher in the experimental group than in the control group. MRI scan showed:the experimental group of the intervertebral disc was better than that of the control group.Conclusion:ADSCs can significantly increaseing the concentration of the type II collagen,proteoglycan and water of the degeneration of intervertebral discafter after the ADSCs transplantation, beside it can increaseing the height of intervertebral space.DAPI can clearly show the ADSC cells after it was implanted the intervertebral disc. |
