Charpter1Chemical&anti-tumor activity research of five kinds of Hainan Polyalthia BL. plantsObjective:To find new active compounds or fraction in Polyalthia BL. plants by systematic chemical separation according to the anti-tumor activity.Methods:Using natural products chemical methods&cytobiology methods to make basic material research of the anti-tumor activity of five Hainan Polyalthia BL. plants.1、The extacts was separated by systematic chemical methods. The extract was mixed with water, extracted by petroleum ether, chloroform, ethyl acetate, respectively. The four fractions were respectively subjected to column chromatography over macroporous resin, normal and reversed-phase silica gel, sephadex LH-20, preparation thin-layer chromatography and recrystallization repeatedly to obtain pure compounds. Their structures were established on. the basis of extensive spectroscopic (NMR、UV、IR、MS) data and by the comparison with reported data in the literature.2、Using MTT assay of cytobiology, test the anti-cancer activity of samples on four common clinical human cancer cell line:SPC-A-1, BEL-7402, SGC-7901&K562.Results:1、24compounds were obtained from the root of P. lauri&the stem of P. rumphii. Among this, compounds9、12、16are novel compounds, separately named as polyalimide A、rumphiin、polyalthurea. Other21are known compounds, isolated from the plants for the first time.2、The anti-cancer activity assay towards the stems,roots&leaves of five kinds of Hainan Polyalthia BL. plants show that the root of P. lauri&the stems,roots&leaves of P. rumphii have strong inhibitory activity towards four cancer cell lines, and the IC50are all under100μg/mL. 3、The anti-cancer activity assay towards the petroleum ether, chloroform, ethyl acetate fractions of root of P. lauri&the stem of P. rumphii shows that the active part is not limited to certain fraction. That indicates the separation work should carry out towards the three fractions separately.4、One novel compound was obtained from the chloroform fraction of P. lauri which shows certain anti-cancer activity with IC50of94.5μg/mL、96.76μg/mL&62.73μg/mL towards SPC-A-1、SGC-7901&K562. Two new compounds were obtained from P. rumphii, one is Rumphiin from chloroform fraction, shows weak inhibitory activity with IC50of187.μg/mL&63.2μg/mL towards SPC-A-1&K562; the other is Polyalthurea from petroleum ether fraction, shows no activity towards the four human cancer cell lines.5、The anti-cancer activity assay towards21known compounds shows that Kolavenic Acid&2-oxo-kolavenic Acid obtained from petroleum ether fraction of P. laui have the most strong inhibitory activities towards the four human cancer cell lines, and the IC50are all less than50μg/mL; and the Oxostephanine obtained from the ethyl acetate fraction of P. rumphii has strong inhibitory activities toward SPC-A-1&BEL-740with IC50of1.47μg/mL&1.73μg/mL separately.Conclusion:The research illustrates the basic material foundation of anti-cancer activity of P. lauri&P. rumphii, the in-vivo activity and the mechanism require further research. Meanwhile, three novel compounds are obtained from the research, that means the there is still big space on the chemical work of Polyalthia BL. plants. Charapter2Preliminary study on in-vivo antitumor activities of Veratrum nigrum L.(Liliaceae)Objective:To study the in-vivo activity of the extracts of Veratrum nigrum L.(Liliaceae) towards the pancreatic cancer cell line SW1990.Methods:Set up the model of human pancreas cancer xenografts in nude mouse, and divide into control group,5-FU group, three dose groups of extracts of Veratrum nigrum L. randomly. The activity was evaluated by the weight anti-tumor ratio&relative volume anti-tumor ratio.Results:Compared with blank control group, the inhibition ratios for increase in xenograft tumor of V. nigrum extracts were14.79%,32.02%and32.96%at oral dose levels of3.75,7.5,15mg/kg for14consecutive days oral dosing, respectively. For5-Fu, positive control group, the inhibition ratio was41.73%after7consecutive days i.p. dosing at15mg/kg. At three dose, the volumn increase of tumor is slowed down, and the relative volume anti-tumor ratio is46.85%、53.43%and47.40%. For5-Fu, positive control group, the inhibition ratio was61.92%after7consecutive days i.p. dosing at15mg/kg.Conclusion:V. nigrum extract inhibits the growth of the xenografts of human pancreatic carcinoma in nude mice and further work would be required to characterize the mechanisms.
