ObjectiveBlood stimulants are stimulants designed to enhance the body’s ability to transport blood oxygen.Athlete Biological Passport(Athlete Biological Passport,ABP)is a long-term record of an athlete’s physiological indicators.Abnormal changes indicate possible doping violations.ABP blood module is an effective means to indirectly detect blood doping through whole blood analysis technology,which greatly improves the detection efficiency of blood doping.However,existing ABP blood module indicators are inadequate,not sensitive to microdose recombinant human erythropoietin(recombinant human erythropoietin,rhEPO),and susceptible to anoxia,dehydration and other factors,so it is necessary to explore new indicators to improve the specificity and sensitivity of ABP blood module.The purpose of this study was to explore the feasibility of ALAS2 mRNA as a new indicator of the blood module of athletes’ biological passport,and to provide a new direction for doping detection.MethodsTwenty healthy male subjects and 18 female subjects were randomly divided into rhEPO experimental group male(E group-M,n=15),rhEPO experimental group female(E group-f,n=13),and saline control group male(C group-M,n=13).n=5)and saline control women(C group-F,n=5).Each subject in the rhEPO group received two doses of 50IU/kg rhEPO.Each subject in the normal saline control group received two injections of normal saline equal to 50IU/kg rhEPO,48 hours apart.18 days before the first injection(day-18),immediately before the first injection(Day 0),24 h after the first injection(Day 1),24 h after the second injection(Day 3),72 h after the second injection(Day 5),1 week after the second injection(day-10),and 2 weeks after the second injection(Day-16)and blood samples were collected 3 weeks after the second injection(Day 23)and 4 weeks after the second injection(Day 30).The blood samples were measured by the whole blood analyzer,including red blood cell count(Red blood cell count,RBC),hemoglobin concentration(Haemoglobin,HGB),hematocrit(Haematocrit,HCT)and other routine indicators of ABP blood module.RNA was extracted from blood samples,and the expression levels of ALAS2 L mRNA and ALAS2L+C mRNA were detected by reverse transcription and real-time fluorescence quantitative PCR(q RT-PCR).Results(1)Two injections of 50IU/kg rhEPO could cause a significant increase in ABP blood module index RET% and IRF.(2)Two injections of 50IU/kg rhEPO could cause a significant decline in the blood module OFFS of ABP.(3)The individual variation coefficient of ALAS2 mRNA expression level in human blood was large.(4)The expression levels of ALAS2 L mRNA and ALAS2 L+C mRNA were significantly increased after two doses of 50IU/kg rhEPO,and the changes of ALAS2L+C mRNA were more significant(5)ALAS2 mRNA was more sensitive to rhEPO than the conventional indices of ABP blood module.Conclusion rhEPO can significantly increase the expression levels of ALAS2 L mRNA and ALAS2 L+C mRNA.ALAS2 mRNA is a potential biomarker of ABP blood module.
