Study On The Medication Rules And Network Pharmacology Of The Method Of Cultivating Soil To Produce Gold In The Treatment Of Chronic Obstructive Pulmonary Diseas

Purpose: Based on the drug utilization regularity of COPD treated with the method of reinforcing soil and generating gold in the past five years,the article utilizes network pharmacology to preliminarily explore the action mechanism of the top four high-frequency drugs(Sijunzi Decoction)in the treatment of COPD.Material and method: The clinical literature concerning the treatment of COPD with the method of reinforcing soil and generating gold published in the three major Chinese databases of CNKI,Wan fang Data and VIP in the past five years were retrieved,and the specific drugs in the collected literature were statistically analyzed using SPSS26.0 software and SPSS Modeler 18.0.The results were discussed.The active chemical components and corresponding targets in Sijunzi Decoction were obtained using TCMSP database.The network diagram of "Chinese medicine-active ingredient-target" was drawn by Cytoscape(3.9.1)software,and the disease targets of COPD were obtained through the joint retrieval of Gene Cards,OMIM,and Dis Gent databases.The intersection targets of Sijunzi Decoction and COPD were obtained using Venny platform,and the core targets were predicted by PPI network diagram.Finally,GO and KEGG analyses were performed using R language.Results:1.The results of medication law of treating COPD by nourishing the soil and generating gold.1.1 High-frequency drugs: Atractylodis Rhizoma,Poria,Glycyrrhrizae Radix,Radix Codonopsis,Radix Astragali,Pericarpium Citri Tangerinae,Rhizoma Pinelliae,Rhizoma Dioscoreae,Radix Platycodonis,Radix Salviae Miltiorrhizae,etc.1.2 The first six kinds of drugs with the most frequent use: tonic drugs,expectorant,antitussive and antiasthmatic drugs,exterior-relieving drugs,heat-clearing drugs,digestion-promoting drugs and qi-regulating drugs.1.3 Results: Most of the drugs are warm,cold and flat;Most of the herbs are sweet,bitter and pungent,and most of them enter the lung and spleen meridians by entering the meridian.1.4 The most supportive drug pairs are Poria-Atractylodes macrocephala,Glycyrrhiza-Atractylodes macrocephala,Codonopsis-Atractylodes macrocephala,Pericarpium Citri Tangerinae-Atractylodes macrocephala,Astragalus-Atractylodes macrocephala,Pinellia ternata-Atractylodes macrocephala,Dioscorea opposita,and Platycodon grandiflorum-Atractylodes macrocephala.1.5 Cluster analysis results: The first category is Amomum villosum,Lablab lablab,Coix seed,Platycodon grandiflorum and Dioscorea opposita,which are the main components of Shenlingbaizhu Powder;The second category is Poria cocos,Atractylodes macrocephala,Pinellia ternata,Pericarpium Citri Tangerinae and Glycyrrhiza uralensis Fisch,which are the main components of Sijunzi Decoction.The third category is Radix Astragali and Radix Saposhnikoviae,which are the main components of Yupingfeng Powder.2.Results of network pharmacology study on treatment of COPD with sijunzi decoction2.1 Sijunzi Decoction may be effective active ingredients in treating COPD,such as porous steroid-7,22-diene-3β-alcohol,7-methoxy-2-methyl isoflavone,etc.The potential core targets are AKT1,TP53,JUN,TNF,CASP3,ESR1,IL6,STAT3 and VEGFA.2.2 The core target of Sijunzi Decoction in treating COPD is involved in steroid hormone response,cell response to chemical stress,drug response,etc.Target protein is mainly distributed in membrane raft,membrane micro-area and membrane area.Molecular functions mainly include G protein-coupled amine receptor activity,nuclear receptor activity and ligand-activated transcription factor activity.2.3 The predictive pathways of Sijunzi decoction in the treatment of COPD mainly include interleukin-17 signaling pathway,small cell lung cancer,chemical carcinogenesis-receptor activation and so on.Conclusion:The drugs for the treatment of chronic obstructive pulmonary disease with the method of reinforcing soil and generating gold are mainly Rhizoma Atractylodis Macrocephalae,Poria,Radix Glycyrrhizae,Radix Codonopsis,Radix Astragali seu Hedysari,and Pericarpium Citri Reticulatae.It is found that the main drugs for tonifying deficiency in the treatment of COPD are the drugs for resolving phlegm,relieving cough and asthma,and the drugs for resolving exterior syndrome,clearing heat,resolving food stagnation,and regulating qi are the secondary ones.The medicinal property of the drug is high with temperature;The five flavors of the medicine are high in sweetness;Lung meridian and spleen meridian were the most meridian tropism of drugs.The six pairs of drugs according to the association rule indicated that tonifying lung and invigorating spleen and resolving phlegm were important methods for the treatment of COPD.The clustering analysis results were similar to those of Shenling Baizhu Powder,Sijunzi Decoction and Yupingfeng Powder.Through network pharmacology,the top four high-frequency drugs(Sijunzi Decoction)in the treatment of COPD may act on such targets as AKT1,TP53,and JUN and participate in various biological processes by virtue of the effective components such as porous steroid-7,22-diene-3β-alcohol,7-methoxy-2-methyl isoflavone and methoxy isoflavone,and exert the effects of anti-oxidative stress response,anti-inflammation,anti-cancer,regulation of cell cycle,and repair of lung tissue mainly through interleukin-17 signal,small cell lung cancer,and chemical carcinogenesis-receptor activation.
