| Objective:To explore the effect and possible mechanism of Guilu Erxianjiao on improving reproductive injury in diabetic rats.Methods:53 SD male rats were randomly divided into 5 groups.1 group was selected as the normal group,and the other 4 groups were combined into the model group.The diabetic model was established by high-fat diet combined with 30 mg·kg-1streptozotocin(STZ)intraperitoneal injection.The model rats were randomly divided into model group,Guilu Erxianjiao group(2g·kg-1),Vitamin E group(0.03 g·kg-1)and Wuzi Yanzong Pill group(0.6 g·kg-1)according to the blood glucose level.The rats were given corresponding drug dose intragastric administration,once a day for 4 weeks,and their body weight and blood glucose were measured weekly.After 4weeks,samples were collected for index detection.Morphological changes of testis and epididymis were observed by hematoxylin-eosin(HE)staining,and apoptosis of testis cells was observed by TUNEL staining.Sperm concentration and motility were detected by semen analyzer.Serum levels of follicle stimulating hormone(FSH),luteinizing hormone(LH)and testosterone(T)were determined by enzyme-linked immunosorbent assay(ELISA).The contents of superoxide dismutase(SOD),propylene glycol(MDA),reactive oxygen species(ROS)and glutathione peroxidase(GPXH)in testicular tissue were determined by ELISA.The expressions of Nrf2,HO-1,Bax and Bcl-2 in testicular tissue were detected by Western blot.Results:Compared with normal group,testicular tissue atrophy,spermatogenic tubules decreased,epididymal wall hyperplasia and lumen stenosis were observed in model group.Sperm concentration and motility decreased(P<0.01).Serum T,FSH and LH levels decreased(P<0.01).The contents of ROS and MDA in testis increased(P<0.01),while the contents of SOD and GPXH decreased(P<0.01).Bax expression increased(P<0.01),Nrf2,HO-1,bcl-2expression decreased(P<0.01).Compared with model group,the pathological changes of testicle and epididymis in Guilu Erxianjiao group were improved to some extent.Sperm concentration and motility were increased(P<0.05,P<0.01).Serum T and LH levels increased(P<0.05,P<0.01),while FSH levels showed no significant difference.The contents of ROS and MDA in testis decreased(P<0.01),while the contents of SOD and GPXH increased(P<0.01).Bax expression decreased(P<0.01),Nrf2,HO-1,bcl-2 expression increased(P<0.05,P<0.01).Conclusion:Guilu Erxianjiao can improve the testicular and epididymal lesions of diabetic rats to a certain extent,and increase the sperm concentration and motility of rats.Increase the serum T and LH levels of rats and regulate the secretion function of HPGA;The apoptosis of testicular cells may be inhibited by regulating the expression of Bcl-2 and Bax.By up-regulating the expression of Nrf2 and HO-1,the contents of SOD and GSH-Px were increased,so as to reduce the levels of ROS and MDA,exert the antioxidant effect,and improve the reproductive damage of diabetic rats.It is worth noting that this study only tracked the fasting blood glucose status of rats for 4 weeks,and did not monitor whether there would be fluctuations in blood glucose after gavage.The influence on fasting or random blood glucose under long-term medication needs further experimental demonstration.Hyperglycemia is the root cause of male reproductive injury.Effective control of blood sugar is still the main measure to prevent and treat diabetes and its complications.In the future,the combination of Guilu Erxian gum and hypoglycemic agents can be considered to explore the effect and mechanism of combined therapy on diabetic reproductive injury,so as to provide a certain reference for clinical trials. |
