Objective:To investigate the effect of Red Yeast Rice(RYR)on lipid metabolism in hyperlipidemia model rats,and to explore the mechanism of improving liver lipid metabolism and regulating lipid in hyperlipidemic rats.Materials and Methods:75 SPF male SD rats aged 8 weeks were fed adaptively for 1week,and the rats were divided into 15 blank rats and 60 model rats by random number table method.The blank group was given a general diet,and the other 60 rats were fed a high fat diet.After a total of 28 days,blood was collected from the caudal vein(n=75),centrifuged at4000 r.min-1 for 10 min,and supernatant was taken to determine the levels of TC,TG,LDL-C and HDL-C in serum.The results TC,TG,and LDL-C were elevated to determine whether the mold making was successful,and the required animals were used in this study The rats with successful mold making were randomly divided into 4 groups,with 15 RYR low,medium and high dose groups.Rats in each treatment group were dissolved in 2.5 ml of 0.9%sodium chloride solution,and the model group was given an equal volume of saline daily once a day for 28 d.The normal group was given ordinary diet,and the other groups were still fed with high fat diet.After the last administration,the rats were fasted for 12 hours,body weight was measured,and blood samples were collected from the abdominal aorta after anesthesia.Serum lipids(TC,TG,HDL-c,LDL-c)and liver function indexes(ALT,AST)of each group were detected by automatic biochemical analyzer.HE and oil red O staining were used to observe the pathological changes of liver in each group.The protein expressions of SREBP-lc,LXR,FAS,ACC and CPT1 in liver tissues were detected by Western Blot and immunohistochemical techniques.m RNA expression of LXR,HMGCR,SREBP-lc,FAS,ACC,CPT-1,ACOX1,PPARα,PPARγ and other factors in liver tissues was detected by PCR.Results:1.The results of body mass and liver index of rats suggested that RYR could improve the body mass and liver index of hyperlipidemia rats.2.The results of serum lipids and liver function indicated that RYR decreased the levels of TC,TG,LDL-C,ALT and AST in serum(P < 0.05,P < 0.01),and increased the level of circulating HDL-C in rats(P < 0.01).3.HE staining and oil red "O" staining results suggested: HE staining and oil red O staining results suggested that RYR could improve liver lipid accumulation and liver injury induced by high fat diet.4.The results of Western Blot and immunohistochemistry indicated that RYR down-regulated the protein expression levels of LXR,SREBP-lc,FAS and ACC,and up-regulated the protein expression levels of CPT1(P < 0.05,P < 0.01).5.PCR results indicated that RYR down-regulated the expression levels of LXR,SREBP-lc,FAS,ACC and HMG-Co Am RNA,and up-regulated the m RNA expression levels of CPT1,ACOX1,PPARα and PPARγ(P < 0.05,P < 0.01).Conclusions:1.RYR can regulate the level of fatty acid in the liver by inhibiting the expression of LXR/SREBP-1c/FAS and increasing the metabolism of fatty acids,thus reducing blood lipids and protecting the liver.
