| Objective:To investigate the ocular demodex killing effect of two aromatic essential oils of gardenia and artemisia argyi in different compatibility ratios,and to determine the safety of their local use.Methods:1.In vitro mite killing experiment:Patients with scales attached to the root of eyelashes and who have not been treated are selected as provide mite volunteers.100%essential oil of gardenia and 100%essential oil of Artemisia argyi were prepared according to 9different compatibility ratios,and the in vitro insecticidal experiment was carried out by immersion method.The changes,survival time and death amount of essential oil of Gardenia-Artemisia argyi were observed and recorded after the action on ocular demodex,so as to determine the insecticidal effect.2.Eye stimulation experiment:30 New Zealand rabbits were randomly divided into 10 groups,including 9 groups with Gardenia-Artemisia argyi essential oil compatibility ratio and tea tree oil positive control group,with 3 animals in each group,and eye stimulation was observed by self-control method.3.Skin irritation experiment:36 guinea pigs were randomly divided into 9 groups,including 9 groups with Gardenia-Artemisia argyi essential oil compatibility ratio,with 4 animals in each group.Self-control method was used to observe the irritation of intact and damaged skin after single or multiple administration of the tested drug.4.Local skin sensitization experiment:18 guinea pigs were randomly divided into 3 groups:Gardenia-Artemisia argyi essential oil compatibility ratio group 1:5,group 2:5 and paraffin oil blank control group,with 6 animals in each group.The allergic reactions of animals in each group were observed and compared after repeated application.Results:1.The activity of demodex could be aggravated by the combination of Gardenia-Artemisia argyi essential oil.In group 0:1,all demodex mites could be killed within10 minutes.Groups 1:5,2:5,2:3,5:2 and tea tree oil could kill all demodex mites within 20min.Groups 1:1,3:2 and 5:1 could kill all demodex within 30min.In group 1:0,all demodex mites were killed within 120min.Within 5 min group 1:5 demodex mortality of 52.17%,higher than21.74%mortality rate of tea tree oil group(?~2=4.572,P<0.05).Within 5 min group 2:5 and2:3 demodex mortality of 52.38%,higher than 21.74%mortality rate of tea tree oil group(?~2=4.454,P<0.05).Within 5 min 1-0 demodex mortality of 77.27%,higher than 21.74%mortality rate of tea tree oil group(?~2=13.878,P<0.05).2.The combination of Gardenia-Artemisia argyi essential oil can cause certain eye irritation.The score of eye irritation in group 0:1 and tea tree oil group was≥4 at 2 hours after treatment,showing mild irritation.Slight edema or slight hyperemia were observed in groups 1:0,1:5,2:5,2:3,1:1,3:2,5:2 and 5:1 at each time,but the score of eye irritation<4 was no irritation according to the scoring criteria.3.No erythema and edema were observed in the complete skin stimulation experiment and the single skin stimulation experiment of damaged skin in the groups of Gardenia-Artemisia argyi essential oil compatibility,and the skin stimulation score was<0.5,showing no irritation.In multiple skin stimulation experiments of damaged skin,1h after treatment,the skin stimulation scores of groups 1:0,2:3,3:2,0:1 and 1:1 were within the range of 0.5～2.99,showing mild irritation.24h after treatment,the skin irritation scores of groups 1:0 and 1:1 were in the range of 0.5～2.99,showing mild irritation.The other groups showed no irritation.4.No allergies or other abnormal reactions were observed in group 5:1,group 5:2 and blank control group on the day of sensitization exposure experiment,day 7 and day 14.No allergic reaction or systemic reaction was observed in group 5:1,group 5:2 and blank control group after excitation and 24,48,72 hours,and the allergy rate was 0%.Conclusion:1.Gardenia-Artemisia argyi essential oil had good acaricidal effect in all the groups of compatibility ratio,among which the acaricidal effect of group 1:5,2:5,2:3 and 0:1 in5 min was better than that of tea tree oil in positive control group.The killing effect of 100%gardenia essential oil and 100%Artemisia argyi essential oil on demodex mites was antagonistic.The killing effect of 100%Artemisia argyi essential oil used in combination was not as good as that of 100%Artemisia argyi essential oil used alone.2.Eye irritation in the group 0:1 and group tea tree oil showed mild irritation to no irritation,while the other groups showed no irritation.3.Gardenia-Artemisia argyi essential oil combination groups showed no irritation to intact skin,but showed mild irritation to damaged skin in groups 1:0,2:3,3:2,0:1and 1:1,and no irritation to other groups.4.The groups 1:5 and 2:5 of Gardenia-Artemisia argyi essential oil compatibility have no sensitization to skin,which has the value of clinical application. |
