Effect Of Simo Decoction On Apoptotic Factors Caspase-3/-9 And NT-pro BNP In Myocardial Cells Of Rats With Chronic Heart Failur | | Posted on:2024-04-02 | Degree:Master | Type:Thesis | | Country:China | Candidate:X F Yin | Full Text:PDF | | GTID:2554307100453264 | Subject:Integrative Medicine | | Abstract/Summary: | PDF Full Text Request | | Objective: To explore the effect of Simotang decoction on the apoptosis factors of cardiac myocytes,caspase-3,caspase-9 and NT-pro BNP in rats with heart failure,to evaluate the therapeutic effect of this drug on chronic heart failure,analyze the possible target of action of Simo Decoction,and provide a new idea for clinical improvement of digestive tract symptoms of heart failure.Methods: A total of 40 male SD rats were randomly divided into a blank group(n=10)and a modeling group(n=30).The heart failure model of rats was established by intraperitoneal injection of isoproterenol,2.5 mg/kg/d,for a total of 4 weeks.The blank group was intraperitoneally injected with physiological saline of the same volume.Twenty four rats with heart failure who were successfully modeled were randomly divided into a model group(n=8),a Simotang group(n=8),and a western medicine group(n=8).With the blank group,this experimental group was divided into four groups.The dosage of Simo Tang oral liquid was 7.56ml/kg/d for the Simo Tang group,6.75mg/kg/d for the Western medicine group,The model group and blank group were gavaged with equal volume of physiological saline for a total of 2 weeks.After the experiment,observe the general situation of the rats in each group.Use ELISA to detect the serum NT pro BNP,HE staining to observe the pathological changes of myocardial cells,Tunel method to detect myocardial cell apoptosis,and Western Blot method to detect the expression of caspase-3 and caspase-9 proteins.Results:1.General state of rats: compared with the model group,both the simotang group and the western medicine group can improve the symptoms of anorexia,edema and fatigue in rats with heart failure to a certain extent;Compared with the western medicine group,the general state of rats in the two groups was not significantly different.2.NT-pro BNP level: compared with the blank group,the content of NT-pro BNP in serum of rats in the model group increased significantly(P<0.05);Compared with the model group,the content of NT-Pro BNP in serum of rats in the Simo decoction group and the western medicine group decreased(P<0.05);Compared with the Western medicine group,the Simo Tang group showed that captopril reduced the serum NT-Pro BNP content in rats,which was better than the Simo Tang group(P<0.05).3.Observation of myocardial tissue HE staining: compared with the blank group,the other three groups of rats have different degrees of myocardial injury,and the order of the degree of injury is:model group>simotang group>western medicine group.4.Myocardial cell apoptosis: The apoptosis rate of the model group was significantly higher than that of the blank group(P<0.05),The apoptosis rate of the Simo decoction group and the western medicine group was lower than that of the model group(P<0.05),while the apoptosis rate of the western medicine group was lower than that of the Simo decoction group(P<0.05).5.Effect on myocardial apoptosis factor: Compared with the blank group,the level of caspase-3and caspase-9 protein in the model group increased significantly(P<0.05);Compared with the model group,the level of caspase-3 protein in Simotang group was lower than that in the model group(P<0.05),caspase-9 protein level had no significant difference compared with model group(P>0.05);Compared with the Western medicine group,the Simo Tang group showed better inhibition of caspase-3 and caspase-9 protein expression by captopril compared to the Simo Tang group(P<0.05).Conclusion:1.Simotang decoction can alleviate the pathological damage of myocardial tissue in rats with chronic heart failure,reduce the rate of myocardial cell apoptosis and the level of serum NT-pro BNP,and evaluate its effectiveness in improving heart failure.2.Simotang can improve heart function and reduce myocardial cell apoptosis in chronic heart failure rats to a certain extent.Part of the mechanism may be related to the down-regulation of the expression of caspase-3 in the apoptosis pathway,but it has little relationship with the expression of caspase-9 in the apoptosis pathway. | | Keywords/Search Tags: | Simotang, Heart failure, Cell apoptosis, caspase-3, caspase-9, NT-Pro BNP | PDF Full Text Request | Related items |
