Objective:To use statistical analysis software to find out the rule of prescription in treating nocturnal enuresis(deficiency of spleen and kidney type)in clinical literature,and to explore its potential core drug prescription through complex network analysis,combined with network pharmacological method to predict its treatment of nocturnal enuresis(deficiency of spleen and kidney type)-related mechanisms.Methods:1.Data mining-based composition rules and new recipe creation:China National Knowledge Infrastructure(CNKI),Wanfang Date Knowledge Service Platform(WF)and China Science and Technology Journal Database(VIP)were searched,the search time was from January 1,2000 to December 30,2022.The clinical literature on the treatment of nocturnal enuresis(deficiency of spleen and kidney type)by Chinese medicine was collected,and the database was established by Excel software,The ancient and modern medical records cloud platform software(personal version v2.3)was used to perform frequency analysis,cluster analysis and association analysis on the drugs extracted from the database;2.Study on the functional mechanism of the new prescription based on network pharmacology:Using HERB Database and literature search,compound of core medicine,based on the Swiss ADME database and the pharmacokinetics of compounds(ADME),potential active compounds were screened out,and the potential targets were predicted by the Swiss Target Prediction database.The disease-related targets were obtained by searching and screening in the disease database.The UniProt protein database was used to annotate the chemical composition targets of the core traditional Chinese medicine,and the genes matching the disease targets were obtained.The corresponding drug effective chemical component target network diagram and core drug key chemical component intersection target disease network diagram were constructed by Cytoscape 3.8.0 software.Then the intersection targets of drugs and diseases are introduced into string 11.5 online platform to construct the protein-protein interaction network diagram,and the obtained protein-protein interaction network diagram is topologically analyzed by using Cytoscape 3.8.0 software to obtain the key targets.Finally,GO analysis and KEGG pathway analysis were carried out for the intersection targets of drugs and diseases by using R software and related installation packages.Results:1.Data mining research:a total of 57 prescriptions were screened,involving 70 traditional Chinese medicines,the total frequency of drug use was 633 times,the 18 most frequently used herbs were cuttlebone.Based on the research results of data mining,it is concluded that Ootheca Mantidis,Alpiniae Oxyphyllae,Cuscutae Semen,Atractylodes macrocephala,Lindera aggregata,Astragalus,Yam and other drugs are mainly used in the treatment of nocturnal enuresis(deficiency of spleen and kidney type),the four Qi are mainly flat,warm and mild,and the five flavors are mainly sweet,pungent and bitter,mostly from the three Zang organs of kidney,spleen and lung;2.Network pharmacology research:There are 81 active ingredients including Menthone,Yakuchinone-A,Tuberostemonine,etc and 22 key targets include ALB,FOS,SLC6A4,IL 6,NTRK2,HTR2A,SRC and so on.There were 1275 items in GO analysis and 34 pathways in KEGG pathway analysis,they are involved in many signaling pathways,such as ligand-receptor interaction pathway,calcium signaling pathway,cAMP signaling pathway,cholinergic synaptic pathway and vascular smooth muscle contraction pathway and other on in the treatment of nocturnal enuresis(deficiency of spleen and kidney type).Conclusion:Based on the data mining results,we conclude that Ootheca Mantidis,Alpiniae Oxyphyllae,Cuscutae Semen,Atractylodes macrocephala,Lindera aggregata,Astragalus,Yam are the main drugs in the treatment of nocturnal enuresis(deficiency of spleen and kidney type),it is characterized by the combination of tonic and astringent drugs.Network pharmacology predicts that the new prescription which is composed of core medicine may be made by Alb,FOS,SLC6A4,IL6,NTRK2,HTR2A,SRC,and so on,it may play an important role in the treatment of nocturnal enuresis(deficiency of spleen and kidney type)by taking part in the interaction of nerve active ligand-receptor,calcium signal pathway,cAMP signal pathway,cholinergic synapse,vascular smooth muscle contraction pathway and so on.
