Purpose:Based on modern literature mining and network pharmacology to study the medication rule of type 2 diabetes complicated with hypertension(DH),further explore the target,signaling pathway and effect of traditional Chinese medicine compound in the treatment of type 2 diabetes complicated with hypertension Mechanism,to provide reference for traditional Chinese medicine screening and further experimental research in the treatment of type 2 diabetes mellitus complicated with hypertension.Method : Relevant materials were retrieved from literature databases such as China National Knowledge Infrastructure(CNKI),CBM,Wanfang,etc.by computer,and the retrieval time limit was set from January 2007 to February 2022.After screening out the clinical efficacy observation of T2 DM combined with hypertension,the literature research database was created,and the data was collected to explore the rules of compound prescription and core drugs.In the database of Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP)database,the drug targets and disease targets of the active ingredients of traditional Chinese medicine compounds were screened out,and the pharmacological information such as the active ingredients of traditional Chinese medicine compounds was screened,and Cytoscape 3.7.1 was used to construct a visualized "drug-target-disease" network.and comprehensive analysis.The intersection targets were imported into the STRING platform to draw the PPI network,and Metascape was used to perform GO analysis and KEGG signaling pathway analysis on the relevant key targets.The DAVID database was used to analyze the GO and KEGG signaling pathways of key targets,and the mechanism of action of traditional Chinese medicine in the treatment of DH was deduced.Results:1 Mata mining resultsThis study strictly followed the inclusion and exclusion criteria and collected a total of73 valid literatures.The system extracted 80 prescriptions for the treatment of type 2 diabetes and hypertension,a total of 109 traditional Chinese medicines,of which 34 traditional Chinese medicines appeared more than 10 times,including Shengdi,Uncaria,Achyranthes,Astragalus,Salvia,etc.The frequency of the four qi has a total of 851 times,the proportion of coldness is the largest in 317 times,and the temperature is the second;The results showed that the liver meridian had the largest proportion of 541 times,the kidney meridian was the second largest,354 times,and the spleen meridian was 273 times.When the support and confidence levels are set to "60%" and "0.8",there are 57 groups of traditional Chinese medicines with a frequency > 80 in all the treatment of type 2 diabetes complicated with hypertension,including 14 medicines,and the core medicines are searched out by streamlining the drug network.To "Birthplace-Uncaria"..The number of matching prescriptions is set to the correlation degree of 6 and the penalty degree of 2 to obtain the degree of association between the two traditional Chinese medicines.The Result is set to 4.The auxiliary platform automatically analyzes 10 groups of potential core compound combinations with high correlation degree.Match,get 5 new recipe combinations: Tianma,Chuanxiong,Turtle,Taizishen;Scutellaria,Mulberry,Keel,Gardenia,Prunella;Polygonatum,Achyranthes,Uncaria,Atractylodes;Pinellia,Chenpi,Uncaria,Atractylodes;Lycium barbarum,ocher,Rehmannia glutinosa,Taizishen.2 Network pharmacology research Results The active ingredients of the core compound "Rehmannia glutinosa,Uncaria,Achyranthes,Astragalus,Salvia" were searched in the TCMSP database,and the corresponding active ingredients and targets were obtained by screening with OB ≥ 30% and DL ≥ 0.18,respectively(2,25),(33,179),(12,162),(20,179),(65,108).Taking Type 2diabetes mellitus" and "Hypertension" as the subject words,673 potential targets of DH were retrieved from TTD and Gene Cards databases,and 673 potential targets of DH were obtained.Using Cytoscape3.7.1 to screen out 89 active ingredients of traditional Chinese medicine compound,among which the active ingredients include quercetin(MOL000098),kaempferol(MOL000422),β-sitosterol(MOL000358)with degree values of 61,25,11.These active ingredients may play an important role in the treatment of DH with traditional Chinese medicine.Using STRING database and R Project software,according to the Count value screening,the top 20 key targets of traditional Chinese medicine in the treatment of DH were listed for visual analysis,and exported with Cytoscape3.7.1 The target of the compound components,it is concluded that the core targets are mainly IL6,TNF,AKT1,IL1 B,VEGFA,CCL2,TP53,PTGS2,GASP3,MMP9,etc.,suggesting that the above targets have a major role in the mechanism of Chinese medicine compound treatment of DH.Gene GO results show that: there are 2013 items P<0.05 in biological process(BP),including reactive oxygen species metabolic process,response to oxygen levels,response to reactive oxygen species Response to reactive oxygen species,regulation of tube diameter,etc.There are 36 items P<0.05 in Cellular Processes(CC),involving membrane rafts,membrane microdomains(membrane rafts)microdomain),etc.There are 43 entries in Molecular Function(MF)with P<0.05,including cytokine activity,heme binding,etc.KEGG enrichment mainly includes AGE-RAGE,HIF1,TNF,IL-17,Fluid shear stress and atherosclerosis and other signaling pathways.Conclusion:1.Clinical research The application of traditional Chinese medicine compound in the treatment of type 2 diabetes mellitus with hypertension is mainly based on nourishing qi and promoting blood circulation,nourishing yin and calming the liver,which is in line with the basic pathogenesis of DH.Most of the medicines return to the liver,kidney and spleen.The nature and taste of medicines are mostly bitter-cold medicines and sweet and warm.Apply the association rule mining method and simplify the drug network to find out the core compounds:Shengdi,Uncaria,Achyranthes,Astragalus,Salvia.Through entropy cluster analysis,5groups of new prescription core combination were obtained,which can be used according to the evidence,and clinical experiments are needed to verify the effect of treating DH.2.Based on the network pharmacology method,the potential mechanism of the core drugs of the traditional Chinese medicine compound "Rehmannia glutinosa,Uncaria,Achyranthes,Astragalus,and Salvia" in the treatment of DH was obtained: quercetin(MOL000098),kaempferol(MOL000422),β-guinea Sterols(MOL000358)are the main components of core drugs,etc.IL6,TNF,AKT1,IL1 B,VEGFA,CCL2,TP53,PTGS2,GASP3,MMP9,etc.act on the targets in the PPI network,by regulating AGE-RAGE,HIF1,TNF,Signaling pathways such as IL-17,Fluid shear stress and atherosclerosis may play a role in the inflammatory response,cellular metabolism,immune stress and other responses of DH patients.
