Objective The purpose of this study is to screen differentially methylated genes in long cancer by targeted bisulfite sequencing(TBS),which is expected to provide valuable biomarkers for early screening,auxiliary diagnosis and even prognosis monitoring of lung cancer.Methods On the basis of the genome-wide DNA methylation profiles and bioinformatics analysis results obtained from 10 pairs of lung cancer tissues and para-cancerous tissues by using RRBS technology in the early stage.In this study,TBS-Panel sequencing was used to detect the methylation levels of specific DMRs in 65 groups of lung cancer tissues and para-cancerous tissues,15 groups of benign pulmonary nodule tissues and para nodular tissues,and the plasma of corresponding patients.Furthermore,the correlation between clinicopathological data(BMI index,sex,smoking history,age,tumor size,tumor stage,turnor pathological type)and the level of CpG island methylation was analyzed.Results 1.In this study,methylation analysis was performed on 160 tissue samples and 30 corresponding patient plasma samples.Combined with the results of stratified integration(BMI index,sex,smoking history),it was found that the CpG island of Forkhead Box Q1(FOXQ1)promoter was at a high level of methylation.And the methylation level of FOXQ1 gene was higher in males and smokers than in females and non-smokers(P<0.001).2.The differences of FOXQ1 gene were analyzed in groups(age,tumor size,tumor stage,tumor pathological type).The results showed that the methylation level of FOXQ1 gene was generally positively currelated with age,tumor site,and tumor stage(P<0.001).Furthermore,the methylatiun level of FOXQ1 gene was significantly higher in squamous cell carcinoma than in adenocarcinoma(P<0.001).Conclusion The promoter region of FOXQ1 gene is obviously regulated by DNA methylation,which may he involved in the occurrence and development of lung cancer.
