| Objective To investigate the expression of hsa_circ_0045800 in peripheral blood of patients with Primary Sjogren’s syndrome(p SS)and its role in the occurrence and development of p SS.Objective To investigate the expression of hsa_circ_0045800 in peripheral blood of patients with Primary Sjogren’s syndrome(p SS)and its role in the occurrence and development of p SS.Methods Fifty patients with primary Sjogren’s syndrome(disease group)hospitalized in the Department of Rheumatology of Ningxia General Hospital from June 2018 to June 2021 and 50 healthy subjects(control group)in the same period were selected.The expression of hsa_circ_0045800 in peripheral blood of the included subjects was detected by q PT-PCR.receiver operating characteristic curve(ROC)was established to evaluate the biological diagnostic value of hsa_circ_0045800 for p SS.The expression level of hsa_circ_0045800 in peripheral blood of p SS patients and its relationship with clinical related indicators,diagnosis,treatment and prognosis were analyzed.The difference in the expression of hsa_circ_0045800before and after treatment in the case group was analyzed.Results1.The expression level of hsa_circ_0045800 in peripheral blood of patients with primary Sjogren’s syndrome was higher than that of the control group(Z=-6.361,P=0.000).2.The receiver operating characteristic curve(ROC)showed that the area under the curve(AUC)was 0.865(95%CI=0.791-0.940,P<0.05),the sensitivity was 74%,and the specificity was92%.The expression level of hsa_circ_0045800 had a good diagnostic efficacy for p SS.3.The expression of hsa_circ_0045800 was positively correlated with age ≥55 years old(Z=-2.149,P=0.032),dry mouth(Z=-2.344,P=0.016),decreased hemoglobin(HGB<120g/L)(Z=-2.991,P=0.032),and decreased hemoglobin(Z=-2.991,P=0.016).P=0.003)and p SS patients with hypothyroidism(Z=-2.276,P=0.022).4.The expression level of hsa_circ_0045800 was positively correlated with age(r=0.328,P=0.019),dry mouth(r=0.331,P=0.017)and hypothyroidism(r=0325,P=0.021).And negative correlation with hemoglobin(HGB)(r=-0.435,P=0.001).5.hsa_circ_0045800 was highly expressed in p SS patients with high disease activity score(ESSDAI score ≥5)(Z=-2.053,P=0.040).p SS patients had lung involvement in ESSDAI score(Z=-1.992,P=0.047)and high expression of hsa_circ_0045800 in renal involvement(Z=-2.322,P=0.014).The expression level of hsa_circ_0045800 was positively correlated with ESSDAI score(r=0.656,P=0.000),lung involvement(r=0.422,P=0.002)and kidney involvement(r=0.330,P=0.019).6.In the case group,the expression level of hsa_circ_0045800 after treatment was lower than that before treatment(Z=-2.075,P=0.037),and the difference was statistically significant.Conclusion1.hsa_circ_0045800 may be involved in the occurrence and development of primary Sjogren’s syndrome.2.hsa_circ_0045800 has the potential to be a biomarker for the diagnosis,assessment of disease severity and prognosis of primary Sjogren’s syndrome. |
