| Objective:1.To investigate the clinical effectiveness and safety of the TJQWG(Toujie Quwen Granule)in the treatment of patients with common COVID-19 infection.2.Test the serum of patients with common COVID-19 infection to explore the effect of TJQWG on inflammation factors.3.To explore the efficacy and anti-inflammation effect of TJQWG in treating virus infected mice.Methods:1.To retrospectively compare the changes in basic information,symptoms,inflammatory indicators,coagulation function,imaging,and liver and kidney function indicators of 272 patients in the treatment group treated with TJQWG and western medicine,and evaluate the effectiveness and drug safety.2.Use MSD highly sensitive multi-cytokine detection technology to detect the level of 71 cytokines in the serum of patients with COVID-19.3.After infecting mice with A/PR/8/34(H1N1)influenza virus,evaluate the weight,lung index,lung tissue pathology,m RNA levels of inflammatory factors in lung tissue,and serum inflammatory factor levels of the mice,and evaluate the effect of TJQWG on the treatment of influenza mice and its impact on the level of inflammation.Results:1.Comparison of baseline data such as patient gender,age,and underlying disease(P>0.05)indicates that the two groups are comparable.The improvement of cough symptoms in the group of TJQWG was significantly better than that in the control group.On the 6th day of treatment,the levels of neutrophils and thrombin time in the control group were higher than those in the TJQWG group(P<0.05);On the 12 th day of treatment,the level of lymphocytes in the patients in the group of TJQWG was significantly higher than that in the control group(P<0.05).On the 18 th day of treatment,the level of white blood cells in the control group was significantly higher than that in the TJQWG group(P<0.05);There was no significant difference in serum amyloid protein,C-reactive protein,and blood lactic acid at each time point(P>0.05);There was a statistically significant difference in the complete disappearance of single lung pneumonia between the two groups before and after treatment.The improvement degree of lung CT in the group of TJQWG was significantly better than that in the control group(P<0.05);After treatment,there was a statistically significant difference in the levels of creatinine and lactate dehydrogenase between the two groups(P<0.05),while there was no statistically significant difference in liver function,urea nitrogen,and creatine kinase levels before and after treatment(P>0.05).2.Comparison of baseline data between the two groups of patients(P>0.05)indicates that the two groups are comparable.After treatment cytokines in serum such as TSLP,IL-3,IFN-α,IL-1β,that levels in the treatment group was significantly lower than that in the control group,and the difference was statistically significant(P<0.05).After treatment,the levels of TPO and IL-4 in the control group were significantly lower than those in the treatment group.The remaining 65 factors had their own trend of change after treatment,but there was no statistical difference between the groups after treatment(P>0.05).3.Compared with the normal group,the lung index in the model group was significantly higher(P<0.05);There was no significant difference in the levels of white blood cells,red blood cells,and procalcitonin(P>0.05);The levels of neutrophils and monocytes increased significantly(P<0.05);The level of lymphocytes decreased significantly(P<0.05);In lung tissue after IFN-α,IL-1β,MCP-1,MIP-1β,RANTES,IL-4,IFN-γ m RNA levels were significantly increased(P<0.05);The other cytokines was no statistical difference in the changes(P>0.05),the levels of IL-6,IL-1β,MCP-1,MIP-1β increased significantly(P<0.05).Compared with the model group,the group of TJQWG has an improvement effect on the weight loss of mice,which can alleviate the pathological changes of viral pneumonia.TJQWG has a reducing effect on lung index(P<0.05);The level of lymphocytes in the group of TJQWG increased(P<0.05);TJQWG effectively reduces the level of TNF-α,IL-1β(P<0.05).There was no statistical significance for other factors(P>0.05);TJQWG can reduce IL-1β,MCP-1,MIP-1β the difference was statistically significant(P<0.05).Conclusion:1.TJQWG can remission the cough,pulmonary inflammatory and lymphocyte level in COVID-19 that can consideration to both effectiveness and safety.2.TJQWG can reduce the levels of pro-inflammatory factors such as TSLP,IL-3,and IL-1 βalso enhance the anti-inflammation level of IL-4 which has a two-way regulatory effect on cytokines and can prevent excessive inflammatory reactions from causing damage to the body.3.TJQWG significantly improves the decrease of lymphocytes in virus infection mice and can reduce TNF-α,IL-1 β,MIP-1 β and MCP-1,which can effectively improve the lung inflammation and the quality of life. |
